Objective: Patients with cutaneous squamous cell carcinoma(CSCC),Bowen's disease(BD),actinic keratosis(AK),and patients with intraocular fistula have normal margins.The tissue was used as the research object to detect and observe the expression of human ?-defensin-2(h BD-2)and eukaryotic transcription factor kappa Bp65(NF-? Bp65).To investigate the differential expression and significance of NF-?Bp65 and h BD-2 in CSCC,BD and AK,and to study the correlation between NF-?Bp65 and h BD-2 in the above diseases.Methods: We used to stained NF-?Bp65,h BD-2 in 10 patients with cutaneous squamous cell carcinoma,10 patients with Bowen's disease,10 patients with actinic keratosis and 10 normal control,and detect their expression levels by immunohistochemical technique(streptavidin-perosidase,SP)and hematoxylin and eosin(HE)staining.Statistical analysis was performed using statistical methods such as Kruskal-Wallis H test,Fisher's accuracy test and Spearman rank correlation using SPSS 21.0 for Windows statistical software package.Results: 1.Expression of h BD-2: There was no expression of h BD-2 in the cytoplasm of the squamous epithelial cells in the normal skin tissue group and the actinic keratosis group;In patients with Bowen's disease and cutaneous squamous cell carcinoma,the expression of h BD-2 in the epithelial cells was diffusely positive.The positive rate of h BD-2 expression in Bowen's disease and cutaneous squamous cell carcinoma was higher than that of normal skin tissue(negative control)(p<0.008);The positive rate of h BD-2 expression in Bowen's disease group and cutaneous squamous cell carcinoma group was higher than that of actinic keratosis group(p<0.008).The positive rate of h BD-2 expression in Bowen's disease and cutaneous squamous cell carcinoma was not statistically significant compared with p=1.00>0.008.2.Expression of NF-?Bp65: In normal skin tissue and actinic keratosis patients,p65 expression was not observed in the nucleus of squamous epithelial cells,and cytoplasmic scatter was positive;In the Bowen disease group and the cutaneous squamous cell carcinoma group,a large amount of p65 was expressed in the nucleus of the squamous epithelial cells,and the cytoplasm was diffusely positive.The expression of NF-?Bp65 in Bowen's disease and squamous cell carcinoma was higher than that in normal skin tissue(negative control)(p<0.008);The expression of NF-?Bp65 in Bowen's disease and squamous cell carcinoma was not significantly higher than that of p=0.579>0.008.3.Correlation analysis between h BD-2 expression and NF-?Bp65 expression: The correlation coefficient of h BD-2 and NF-?Bp65 expression in Bowen's disease was rs=0.504,P=0.137,the difference was not statistically significant;The correlation coefficient of h BD-2 and NF-?Bp65 expression in cutaneous squamous cell carcinoma was rs=0.557,P=0.081,and the difference was not statistically significant.It is suggested that h BD-2 expression is associated with the wireless expression of NF-?Bp65 in Bowen's disease and cutaneous squamous cell carcinoma.Conclusions: h BD-2 may be involved in the formation of CSCC.NF-?Bp65 may be involved in the formation of CSCC.
