Background:Pancreatic cancer is an almost uniformly lethal disease which has low early diagnosis rate,rapid progress,poor prognosis.Generally pancreatic cancer is advanced from precancerous lesions of the pancreas,which is widely regarded consisting of pancreatic intraepithelial neoplasia,intraductal papillary mucinous neoplasm,and mucinous cystic neoplasms.TRIP6 is an adaptor protein of structural and functional diversity.It plays a role in various of cancer progression.Objective: This article aims to investigate the role of TRIP6 in the development and progression of pancreatic cancer through observing and analyzing the expressions and clinical significance of TRIP6 in normal pancreatic tissues,precancerous lesions of pancreas tissues and pancreatic cancer tissues.Methods: Collected 151 cases of pancreatic tissues resected in Fuzhou general hospital from January 2012 to December 2016.Including 43 cases of normal pancreatic(NP)tissues,20 cases of intraductal papillary mucinous neoplasm(IPMN)tissues,20 cases of Mucinous Cystic Neoplasms(MCN)tissues,18 cases of(86 foci)pancreatic intraepithelial neoplasia tissues(including 25 foci PanIN-1,33 foci PanIN-2,28 foci PanIN-3)and 50 cases of pancreatic cancer tissues.As well as the clinicopathologic data of pancreatic cancer cases were included.We detected the expressions of TRIP6 in each group through Immunohistochemical SP method.Finally,we quantified immunohistochemistry results using IPP(Image-Pro Plus 6.0)professional imaging analysis system.Statistical analysis was performed using SPSS 20.0 statistical software.And the expressions of TRIP6 in different tissues were analyzed,as well as the correlation between TRIP6 expression and clinicopathological characteristics of pancreatic cancer cases.Results: 1.The expression of TRIP6 was negative or little in normal pancreatic tissues(0.0102±0.0031).There were different degrees of positive expressions of TRIP6 in IPMN(0.0702±0.0201),MCN(0.0851±0.0315),PanIN-1(0.0285±0.0029),PanIN-2(0.0547±0.0135),PanIN-3(0.0896±0.0245)and pancreatic cancer tissues(0.0935±0.0304).2.The expression level of TRIP6 in any different lesion of pancreas tissues was higher than that in normal pancreatic duct tissues(P<0.05).3.The expression level of TRIP6 was higher in pancreatic cancer tissues than that in IPMN tissues(P<0.05).There was no difference between expression level of TRIP6 in pancreatic cancer and MCN tissues(P>0.05).4.The expression level of TRIP6 increased with progression of the lesions in the following order: NP?PanIN-1? PanIN-2?PanIN-3?pancreatic cancer,with statistical significance(P<0.05).5.The expression level of TRIP6 was associated with neural invasion(P<0.001),lymph node metastasis(P=0.013),distant metastasis(P=0.018),differentiation(P=0.019),CA19-9 level(P<0.001)in pancreatic cancer.While the expression of TRIP6 was independent of other clinicopathologic data(P?0.05).Conclusion: The expression of TRIP6 is much higher in pancreatic cancer and precancerous lesions of pancreas than that in normal pancreatic tissues,and it increases with the degree of abnormality of pancreatic tissue.Therefore increasing expression level of TRIP6 is an early event of pancreatic cancer.TRIP6 is involved in the development and progression of pancreatic cancer.Abnormally high expression of TRIP6 may promote neuroinfiltration,invasion and migration of pancreatic cancer.
