Research On Anticoagulant Active Ingredients From Sorghum Root

Abstract/Summary:
Sorghum root is the root of sorghum(Sorghom vulgare Pers.),which is also a traditional Chinese herbal medicine,which plays a significant role in the treatment of hydrocephalus,orthopedic diseases and anti-fatigue.The results showed that the extraction site(BES)and the supernatant(WEAE)of sorghum root alcohol extraction had obvious effect of dilating blood tubules and anticoagulant.On the basis of previous research,the chemical constituents of the active extracts of sorghum root were analyzed comprehensively,and the material basis of the anti-coagulation effect was studied in combination with the activity tracing method The main results are as follows:1.Study on anti-platelet aggregation activity of 16 fractions and 2 extracts in vitro.At a concentration of 100 μg/m L,WEAE,BES-M,BES-M30%,WEAE-M30%,and WEAE-D60% fractions were significantly inhibited collagen-induced platelet aggregation.The low-dose group,BES-M30%,the medium-dose group BES-M30%,and the high-dose group WEAE were significantly inhibited collagen-induced platelet aggregation.Under thrombin-induced platelet aggregation,WEAE-M30%,medium-dose group BES-M30%,and high-dose group WEAE were showed the significantly inhibited platelet aggregation activity.2.The samples in WEAE-M30% and BES-M30% fractions from sorghum root were tested by HPLC.The chromatographic conditions were: injection volume 10μL;chromatographic column Accurasil C18(250 mm × 4.6 mm);wavelength 230nm;Temperature 35℃;flow rate 1 m L/min;mobile phase(A)methanol,(B)0.2%phosphoric acid aqueous solution,elution gradient 0-5 min A: 5%-7%,B: 95%-93%;5-10 min A: 7%-10%,B: 93%-90%;10-15 min A: 10%-15%,B: 90%-85%;15-40 min A:15%-18 %,B:85%-82%;40-45 min A: 18%-20%,B: 82%-80%;45-55 min A: 20%-25%,B: 80%-75%.The results show that the chemicalcomposition of the two groups is different,and there are many chemical constituents in the former.3.Adopted LC/MS to analyze the chemical ingredients of active fractions,namely WEAE-M30% and BES-M30%,which mainly include flavonoid glycosides and phenolic acid compounds.It was inferred that the anticoagulant ingredients in sorghum root were flavonoid glycosides and phenolic compounds.4.Twelve compounds were isolated from sorghum root through column chromatography.Their structures were characterized by MS,1D NMR and 2D NMR data as 4-[(6’-O-β–D-glucopyranosy)oxy] hydroxyphenylacetic acid cyclic dimeric inner ester(1),β-sitosterol(2),stigmastero(3),daucosterol(4),4-methoxycinnamic acid(5),taxiphyllin(6),chlorogenic acid(7),succini acid(8),p-hydroxybenzaldehyde(9),trans-p-hydroxycinnamic acid(10),obtusalin(11).mannitol(12),respectively..Compounds 4-5 and 9-12 were reported from this species for the first time,especially compound 1 was new compounds,named Sorgholide A.Its structural characteristics was 24 ring dimeric double lactonol glycoside formed by reverse polymerization of p-hydroxyphenylacetate glucoside,5.In vitro anti-platelet aggregation activity of compounds was studied,with adenosine diphosphate(ADP),collagen and thrombin as inducers.The results were that Sorgholide A and taxiphyllin were showed inhibit platelet aggregation induced by ADP,collagen and thrombin;trans-p-hydroxycinnamic acid was showed inhibitory activities on platelet aggregation induced by ADP;Succinate was exhibited significantly inhibits ADP and thrombin-induced platelet aggregation.
