Diseases related to acute gastric mucosal lesions,such as gastric ulcer,has an incidence of about 5%to 10%.The patient has stomach pain,flatulence,nausea and vomiting,acid reflux and other gastrointestinal symptoms.Gastric ulcer has a long course and is easy to recur,if it is not properly and timely treated,it will often lead to adverse consequences such as upper gastrointestinal bleeding,ulcer perforation,and pyloric obstruction,bringing threats to life safety of patients.For decades,controlling gastric acid secretion and neutralizing gastric acid have become the basis of ulcer therapy.However,proton pump inhibitors,H2 receptor antagonists and other chemicals commonly used in the treatment of gastric ulcers in clinical practice have adverse side effects.And some researchers have suggested that the treatment target for gastric ulcer should not be limited to controlling gastric acid.A variety of factors can cause acute gastric mucosal damage,alcohol,as a common psychoactive substance of abuse,is generally considered to be one of the factors that cause gastric ulcers.With the development of departments such as molecular biology,the mechanism of the stomach trouble caused by ethanol was gradually disclosed.The mechanism by which ethanol damages gastric mucosa are:infiltration of neutrophils in the gastric mucosa injury site triggers an inflammatory reaction,and leads to an oxidative stress.Evodiamine and curcumin are active ingredients of traditional Chinese medicine that protect gastric mucosa from ethanol.The mechanism by which evodiamine plays a protective role involves improving the antioxidant and anti-inflammatory status of gastric tissues.Curcumin regulates inflammatory cytokines and oxidative stress when it protects gastric mucosa from ethanol-induced damage.Traditional Chinese medicine has a wide range of paths preserving protective effect and little side effects.It can make up for the limitations of Western medicine in treating gastric ulcers,and has advantages in improving the quality of ulcer healing and reducing the recurrence rate.The first part,preparation and physicochemical properties of EJPCN.Objective:To prepare EJPCN and determine EJPCN physicochemical parameters.Method:EJPCN was prepared and its physicochemical parameters were investigated.Results:EJPCN is a clear,blue opalescent liquid with a conductivity of?4.36±0.02??S·cm-1 and a pH of 1.84±0.01.The second part,the protective effect of EJPCN on ethanol-induced gastric mucosal damage.Objective:To study the protective effect of EJPCN on acute gastric mucosal injury induced by absolute ethanol in rats,and to explore its mechanism.Method:The rats were randomly divided into 7 groups,the normal group,the model group,the positive drug group,the drug substance treated group?EVO:CUR,40:20,mg/kg?,and the EJPCN low-dose group?EVO,10 mg/kg?,EJPCN medium-dose group?EVO,15 mg/kg?,EJPCN high-dose group?EVO,20 mg/kg?,8 rats in each group.The positive drug group was orally administered with 50 mg/kg rebamipide,and the drug substance treated group was orally administered with EVO and CUR mixture in 0.5%CMC-Na suspension;10 mg/kg,15mg/kg,and 20 mg/kg?calculated as EVO?of nanoemulsion were administered orally to the low,medium,and high dose groups of EJPCN,and the volume was 1.0 mL/100g·bw,the normal group and the model group were given an equal volume of normal saline once a day,for 14 days.Before dissection on day 15,all animals were fasted but free for water for44 h,and then fasted and no access to water for 4 h.Animals in each group were given 5 mL/kg of ethanol,and normal saline was given 5 mL/kg to normal group.Two hours after modeling,the rats were sacrificed and their stomachs were removed.Observe the gastric mucosal damage with the naked eye,take pictures,count the glandular gastric ulcer index,and store the stomach at-80?for detection of biochemical indicators.Results:Compared with the model group,the activity of SOD in EJPCN-treated group was elevated,and the content of MDA decreased.EJPCN increased the content of PGE2 and IL-4,and reduced the levels of pro-inflammatory factors IL-6 and TNF-?.Conclusion:EJPCN has a certain protective effect on gastric mucosal injury in rats induced by absolute ethanol,which is mainly achieved by antioxidant and inhibiting the release of inflammatory mediators and improving PGE2 protective substances.The third part,effect of EJPCN on body weight and organ index of rats.Objective:To study the effect of EJPCN on body weight and various organ index of rats.Method:During the second part of the pharmacodynamics experiment,the weight of rats was weighed and recorded daily.After the rat was sacrificed,the heart,liver,spleen,lung,kidney and brain of rats were promptly removed with the adhered fat and fascia removed,then was washed with saline and dried with the filter paper,then the mass of the above organs was weighed and recorded,organ index was calculated according to the formula in this article.Results:During the feeding period,the weight of rats in each group showed a gradual increase trend,and the weight gain of rats in the EJPCN middle-dose group was slower than that of other groups.In the previous days,there was no significant difference in the weight of rats in each group.On the sixth day,the body weight of the EJPCN middle-dose group was significantly different from that of the EJPCN high-dose group?p<0.05?,and was significantly different from that of the other groups?p<0.01?.On the 8th and 10th day,the body weight of rats in the EJPCN middle-dose group was significantly different from that of the other groups?p<0.01?.On the 12th day,the body weight of the EJPCN middle-dose group was significantly different from that of the EJPCN low-and high-dose groups?p<0.05?,and the body weight of the EJPCN middle-dose group was significantly different from that of the other groups?p<0.01?.On the 14th day,the body weight of the EJPCN low-dose group was significantly different from that of the normal group and the model group?p<0.01,p<0.05?;the body weight of the EJPCN medium-dose group was significantly different from the other groups besides the EJPCN low-and high-dose groups?p<0.01?;the body weight of the EJPCN high-dose group was significantly different from the normal group and model group?p<0.01,p<0.05?.EJPCN had no significant effect on the organ index of heart,spleen,lung,kidney and brain in SD rats.Compared with the model group,the liver index of EJPCN-treated group was increased.Conclusion:EJPCN has a certain effect of delaying the weight gain of rats,and has a certain effect on liver index.
