OBJECTIVE:To observe the effect of Tanshinone ?A Impacts on the expression of HIF-1 and VEGF in choroid neovascularization(CNV)of animal model,and to explore the intervention effect and mechanism of Tanshinone ?A on CNV.METHODS To choose 60 large male BN rats which age between 7-8 weeks were randomly divided into 6 groups : Blank group(A)10,Model group(B)10,Western Medicine Treatment Group(C)10,Tanshinone ?A low dose group(D)10,Tanshinone ?A in the dose group(E)10,Tanshinone ?A high dose group(F)After the treatment 7,14,21 days,to give Fundus photo,Fluorescein Fundus Angiography(FFA)Indocyanine Green Angiography(ICGA).To use PCR to detect the HIF-1 ? and VEGF expression.RESULTS:(1)CNV central thickness: at 7 days,the thickness of the model group was the thickest,and that of the Lucentis group was the thinnest,compared with that of the Tanshinone group(all P < 0.05);the thickness of the Tanshinone group was between the two groups,compared between the groups(P > 0.05);at 14 days,the thickness comparison trend of each group was the same as that of the 7 days group;(2)CNV formation rate,at 7 days,CNV formation rate increased,reached the peak at 14 days,21 days The formation rate of CNV was over 56.7%.(3)The average optical density of fluorescein leakage was lower in Lucentis group and Tanshinone group than that in model group(P < 0.05)and Tanshinone group(P > 0.05).(4)The results of ELISA showed that at 14 days,the inhibition of VEGF was the strongest in Lucentis group,Compared with the model group and Tanshinone group(all P < 0.05),the expression of VEGF in Tanshinone group was between the two groups(P > 0.05).At 7 days,the inhibition tendency of VEGF in each group was similar to that of 14 days.The results of HIF-1 ? detected by ELISA showed that the inhibition tendency of VEGF was similar to that of ELISA The results of PCR showed that the trend of VEGF and HIF-1 ? was consistent with those of ELISA.CONCLUSIONS:Tanshinone ?A can reduce the expression of HIF-1 and VEGF in choroid retinas of animal models of CNV,thus inhibiting the growth of experimental CNV generated by semiconductor laser photocoagulation in fundus.
