Breast cancer is the most commonly diagnosed cancer type among women all around the world, and more than 90%of patients die from tumor metastasis,so the prevention and treatment of breast cancer is particularly important. In China,a developing country with over a billion people, the screening and treatment of breast cancer is more difficult to carry out,so the incidence and mortality rates of breast cancer in the past a decade remain high. The standard breast cancer treatment strategies include surgery, radiation therapy and drug therapy(chemotherapy, hormone therapy, targeted therapy). For the current domestic market, the traditional anti-cancer drugs are still dominant.Unfortunately, owing to various problems, the clinical use of these drugs is limited. Therefore, the development of new anti-breast cancer drugs is especially urgent.Traditional Chinese Monomer Lycorine,a natural alkaloid, has been reported to exhibit a wide range of physiological effects, including the potential effect against cancer. In our study, the anti-tumor potency of Lycorine on breast cancer was evaluated in vitro and in vivo for the first time. Our results indicated that lycorine inhibited breast cancer cells growth, migration and invasion as well as induced apoptosis. In vivo study, lycorine not only suppressed breast tumor growth in xenograft models, but also inhibited breast tumor metastasis in MDA-MB-231-Luc tail vein model. Moreover,we found that there is no significant toxicity with the dosage of Lycorine.Furthermore, from this respect of mechanism, lycorine inhibited tumor cell migration and invasion via blocking the Src/FAK(focal adhesion kinase)-involved pathway. In conclusion, our study implied that lycorine was a potential candidate drug of breast cancer by inhibition of tumor growth and metastasis.
