Objective: To study and discuss the role of Atg5 and Atg7 in the pathogenesis of polymyositis(PM)and dermatomyositis(DM)patients.Methods: 1.The changes of skeletal muscle fibers in PM,DM and control group were observed by HE staining.2.Envision two-step method was used to detect the expression levels of Atg5 and Atg7 in the muscle tissues of PM,DM patients and control group.3.Correlation analysis between Atg5 and Atg7 immunohistochemistry scores and MYOACT scores in PM and DM groups respectively.Results: 1.According to the microscopic comparison between the PM group,the DM group and the control group under HE staining,incomplete skeletal muscle fibers,atrophy and lymphocyte infiltration were observed in all the PM group and the DM group.2.The positive rate of Atg5 in PM group and DM group was significantly higher than that in control group(P < 0.001);.The positive rate of Atg7 in the PM group and DM group was significantly higher than that in the control group(P < 0.001).3.The immunohistochemical score of Atg5 in the muscle tissues of the PM group and the DM group was positively correlated with the total MYOACT score(correlation coefficient r was 0.802 and 0.699,respectively,all P < 0.05),and the immunohistochemical score of Atg5 was positively correlated with the MYOACT muscle score(correlation coefficient r was 0.748 and 0.767,respectively,all P < 0.05);The immunohistochemical score of Atg7 in the muscle tissues of the PM group and DM group was positively correlated with the total MYOACT score(correlation coefficient r was 0.692 and 0.713,respectively,all P < 0.05),and the immunohistochemical score of Atg7 was positively correlated with the MYOACT muscle score(correlation coefficient r was 0.738 and 0.852,respectively,all P < 0.05).Conclusion: Atg5 and Atg7 could involve in PM/DM by autophagy,which being positively correlated with disease activity.Atg5 and Atg7 are expected to be new indicators for assessing PM/DM disease activity.
