Objective: Myocardial ischemia-reperfusion injury(MIRI)is an unavoidable problem in the treatment of coronary heart disease with reperfusion.Bawei Chenxiang powder is a classic Tibetan medicine used in clinical treatment of coronary heart disease in Tibetan medicine.The material basis is not yet clear.The experiment is to establish a model of myocardial ischemia-reperfusion injury by coronary artery ligation,to study the effect of Bawei Chenxiang Powder on myocardial ischemia-reperfusion injury;liquid-mass spectrometry and gas-phase mass spectrometry are used to analyze the main chemical components Combined with the method of serum medicinal chemistry,the material basis of baweichenxiangsan for protecting myocardial ischemia-reperfusion injury was initially revealed,which provided a scientific experimental basis for clinical application.Methods: 1.The rat model of myocardial ischemia-reperfusion injury was established by ligating the left coronary artery for 30 minutes and reperfusion for 60 minutes.SD rats were divided into a sham operation group,a model group,a low-,medium-,and high-dose group of Bawei Chenxiang Powder,and each group was modeled after the last administration.After the experiment,blood was taken from the abdominal aorta for the detection of LDH and CK.After the blood was taken,the heart was taken out to observe the myocardial tissue morphology,myocardial infarction range,myocardial histopathological changes,and myocardial ultrastructure changes.2.UPLC-Q-Exactive quadrupole-electrostatic field orbital trap high-resolution mass spectrometry and gas chromatography-mass spectrometry were used to detect the chemical components of Bawei Chenxiang Powder in the experiment.Extraction of non-volatile oil components of Bawei Chenxiang San compound by methanol ultrasonic method,used for detection and analysis of chemical components of Bawei Chenxiang San compound non-volatile oil;Extraction of volatile oil components of Bawei Chenxiang San compound by steam distillation,used for chemical analysis of volatile oil of Bawei Chenxiang San compound Detection and analysis of ingredients.3.UPLC-Q-Exactive quadrupole-electrostatic field orbital trap high-resolution mass spectrometry was used in the experiment to detect the components of Bawei Chenxiang Powder scattered into the blood.SD rats were divided into two groups,the blank group and the Baweichenxiang powder medicine group,and the blank group was administered with an equal volume of distilled water by intragastric administration.Based on analysis and comparison of rat serum after administration of Bawei Chenxiang San suspension and Tibetan medicine Bawei Chenxiang San compound extract,the prototype chemical components in the serum samples were initially determined.Results: 1.Protective effect of Tibetan medicine Bawei Chenxiang Powder on myocardial ischemia-reperfusion injury in rats(1)Effect of Bawei Chenxiang Powder on serum lactate dehydrogenase(LDH)and creatine kinase(CK)activity in ischemia-reperfusion ratsEach dose group of Bawei Shenxiang Powder can reduce the serum lactate dehydrogenase(LDH)and creatine kinase(CK)activities of myocardial ischemia-reperfusion injury rats,and the high-dose group has the best effect.(2)Effect of Bawei Chenxiang Powder on the extent of myocardial infarction in ischemia-reperfusion ratsThe middle and high dose groups of Bawei Chenxiang Powder significantly improved the range of myocardial infarction caused by ischemia-reperfusion injury.(3)Effect of Bawei Chenxiang Powder on myocardial histopathology in ischemia-reperfusion ratsMyocardial cell interstitial edema,capillary dilation,and inflammatory cell infiltration in the rats of each dose group of Bawei Shenxiang Powder were all less severe than those of the ischemia-reperfusion model group.(4)Effect of Bawei Chenxiang Powder on myocardial ultrastructure of ischemia-reperfusion ratsCompared with the model group,the low-and middle-dose Bawei Chenxiang Powder group had less damage to the cardiomyocytes;compared with the model group,the structure of the myocardial cells was normal in the high-dose group of Bawei Chenxiang Powder.2.Qualitative analysis of in vitro chemical composition of Tibetan medicine Bawei Chenxiang Powder(1)Qualitative analysis of chemical components of non-volatile oil in vitro from Bawei Chenxiang PowderBy comparing the total ion diagram,mass spectrum,related literature data and database of the extract of Bawei Chenxiang Powder Compound,27 chemical components of nonvolatile oil were initially identified from Bawei Chenxiang Powder Compound.(2)Qualitative analysis of chemical constituents of volatile oil from Bawei Chenxiang PowderBy comparing the GC-MS total ion diagram,related literature data,and database information of Bawei Chenxiang Powder compound extract,40 chemical components of volatile oil were initially identified from Bawei Chenxiang Powder compound.3.Qualitative analysis of blood components of Tibetan medicine Bawei Chenxiang PowderBy analyzing and comparing the in vitro samples of Bawei Chenxiang Powder,the total ion chromatogram of the serum of the administration group,the mass spectrum,the relevant literature data and the database,a total of 16 prototype blood components were identified from the serum of rats administered Bawei Chenxiang Powder.Conclusion: 1.This experiment establishes a model of myocardial ischemia-reperfusion by using coronary ligation,and confirms that Bawei Chenxiang Powder has a positive effect on protecting myocardial ischemia-reperfusion injury in rats,and an anti-myocardial ischemia-reperfusion injury effect.Significantly reduced LDH and CK values in rats,effectively reduced myocardial ischemia-reperfusion injury,significantly reduced the range of myocardial infarction in myocardial ischemia-reperfusion injury rats,and improved cardiac function to a certain extent.2.In this experiment,UPLC-Q-Exactive quadrupole-electrostatic field orbital trap high-resolution mass spectrometry and gas-mass spectrometry were used to determine 67 chemical components of Tibetan medicine Bawei Shenxiangsan(including 27 nonvolatile oil chemical components,40 kinds of volatile oil.)3.Based on the research ideas of serum medicinal chemistry of traditional Chinese medicine,this experiment uses UPLC-Q-Exactive quadrupole-electrostatic field orbital trap high-resolution mass spectrometry to analyze the blood components of Tibetan medicine Bawei Chenxiang Powder scattered,and determine 16 prototype non-The blood-injection components of volatile oil,most of the above blood-injection prototype components have certain anti-inflammatory and anti-oxidative effects and protect the heart from vasodilation.It is speculated that the blood-injection components of these 16 prototype may be effective for myocardial ischemic injury of Bawei Chenxiang Powder Ingredients,preliminary reveals the medicinal substance basis of this medicine,and provides scientific experimental basis for the prevention and treatment of ischemic heart disease by Bawei Shenxiang Powder.
