Purpose:Breast cancer has become a common malignant tumor in women.This topic proposed by in vitro experiment research dandelion flavonoids of MCF-7 cell biology behavior of breast cancer,namely the influence of cell proliferation and apoptosis,aims to address the dandelion flavonoids function and its mechanism of action of breast cancer,to give experimental basis toapplyitin the clinical treatment of breast cancer,and for the future development of new natural medicine to provide new ideas.Methods:In vitro human breast cancer MCF-7 cell,using the CCK-8 method used in the determination of cell proliferation activity at different concentrations(50,100,200,400,600 ?mol/L)of dandelion flavones and different concentrations(25,50,100,200,400 ?mol/L)of 5-fluorouracil(5-FU)on MCF-7 cell proliferation inhibition,and it is concluded that the two drugs respectively 24 hours half inhibitory concentration(half maximal inhibitory concentration,IC50);The effects of dandelion flavonoids and 5-FU on apoptosis of MCF-7 cells were tested by flow cytometry.RT-PCR was used to determine the effect of dandelion flavonoids and 5-FU on the expression of P53,Bax and Bcl-2 m RNA in MCF-7 cells.Results:The IC50 of dandelion flavonoids and 5-FU on MCF-7 cells for 24 h was 253.6 ?mol/L and 105.9 ?mol/L,respectively.Dandelion flavonoids had inhibitory effect on the proliferation of MCF-7 cells,and the inhibitory effect increased with the increase of the concentration(all P <0.05).The inhibitory effect was stronger at 48 h than at 24h(P<0.05).Flow cytometry showed that 250 and 500 ?mol/L of dandelion flavonoids could promote the apoptosis of MCF-7 cells,the apoptosis rate was positively correlated with the concentration,and flavonoids could coordinate with 5-FU to promote the apoptosis.RT-PCR detected that after adding 250 and 500 ?mol/L of dandelion flavonoids with the same concentration of 5-FU,the m RNA expression of P53 and Bax in MCF-7 cells increased with the increase of flavonoid concentration,while the m RNA expression of Bcl-2 decreased with the increase of concentration(P<0.05).Conclusions : Dandelion flavonoids may affect the proliferation and apoptosis of breast cancer MCF-7 cells by regulating the expression of P53,Bax and Bcl-2.
