Objective:To detect the expression levels of miR-10b and Tiam1 mRNA in individual tissues of the same gastric cancer,to analyze how they were expressed and the relationship between them and pathological typing,and to explorewhether miR-10b and Tiam1 mRNA are related to various inflammation indicators during the development of gastric cancer.Methods:A total of 80 samples were collected from 40 patients with gastric cancer confirmed by the Gastrointestinal surgery Department of the First Hospital of Jilin University from December 2017 to December 2018,as well as the corresponding adjacent tissues 5cm from the edge of the cancer tissue.Meanwhile,preoperative whole blood and serum samples of the patients were also collected.The expression levels of miR-10b,Tiam1 mRNA in cancer tissues and adjacent tissuesand the expression levels of miR-10b in serum samples were detected by q RT-PCR,and the contents of C-reactive proteinand Albumin were determined by immunonephelometry and bromocresol green method respectively,and the C-reactive protein-to-albumin ratio was calculated.Systemic immune inflammation index,Platelet-to-lymphocyte ratio,Neutrophil to lymphocyte ratio and Lymphocyte to monocyte ratio were calculated based on routine blood test results.The relationship between miR-10b and Tiam1 mRNA in individual tissues of the same gastric cancer and its pathological typing were analyzed.The correlation between the expression of miR-10b and Tiam1 mRNA in tissues and the expression of miR-10bin serum and various inflammatory indicators was analyzed to measure the correlation.Results:1.The expression level of miR-10b in gastric cancer tissues decreased in72.5% of gastric cancer individuals compared with that in the adjacent 5cm tissues,and the expression level of miR-10b in gastric cancer tissues was significantly lower than that in the adjacent tissues(P< 0.05).The expression level of Tiam1 mRNA in gastric cancer tissues increased in 80% of gastric cancer individuals compared with that in the adjacent 5cm tissues,and the expression level of Tiam1 mRNA in gastric cancer tissues was significantly higher than that in the adjacent tissues(P< 0.0001).2.The expression of miR-10b and Tiam1 mRNA in 27 of the 40 patients with gastric cancer was in line with the targeted relationship between down-regulation of miR-10b and up-regulation of Tiam1 mRNA(67.5%,27/40),of which 24 showed decreased expression of miR-10b and increased expression of Tiam1 mRNA;3showed increased expression of miR-10b and decreased expression of Tiam1 mRNA.Of the 27 patients with gastric cancer in relation to targeted,with TNM staging ? period,low differentiation,mixed type,tumor diameter?5cm,ulcers,infiltration depth has reached serous,no retinal vascular and nerve invasion,infiltration and no cut edge carcinoma.3.Correlation analysis on the expression levels of miR-10b and Tiam1 mRNA in gastric cancer patients showed a significant negative correlation between miR-10b and Tiam1 mRNA(r=-0.331,P<0.05).4.The expression levels of miR-10b and Tiam1 mRNAin different TNM stages of gastric cancer patients showed different trend.The expression level of miR-10b decreased with the progress of the clinical TNM stage of gastric cancer which the expression level ofmiR-10b in patients with ? ~ ? clinical stage was lower than those of ?~?clinical stage.The expression level of Tiam1 mRNAincreased with the progress of the clinical TNM stage of gastric cancer which the expression level of Tiam1 mRNA in patients with ? ~ ? clinical stage was higher than those of? ~ ? clinical stage.The expression level of miR-10b in lymph node metastatic gastric cancer tissues was significantly lower than that in non-metastatic lymph nodes(P<0.05).There was no significant difference between miR-10b and Tiam1 mRNAin age,sex,tumor size,differentiation degree,Lauren typing group and gross morphology(P> 0.05).5.Correlation analysis of the expression levels of miR-10b,Tiam1 mRNA in gastric cancer tissues and inflammatory indicators showed that miR-10b and Tiam1 mRNA were not correlated with SII,PLR,NLR,LMR and CAR,while the expression level of serum miR-10b and PLR were positively correlated(r = 0.428,P<0.05),and were not correlated with SII,NLR,LMR and CAR.Conclusions:1.The expression level of miR-10b is lower in gastric cancer tissues and the expression level of Tiam1 mRNAis higher in gastric cancer tissues,and correlation analysis showed a significant negative correlation between the expression levels of miR-10b and Tiam1 mRNA.It suggested that the down-regulation of miR-10b and the up-regulation of Tiam1 mRNA in gastric cancer may be one of the pathogenesis of gastric cancer.2.The correlation between the expression levels of miR-10b,Tiam1 mRNA and the clinicopathological characteristics of gastric cancer patients suggests that the expression levels of miR-10b and Tiam1 mRNA can reflect the progression of gastric cancer and have important clinical value.3.The expression level of miR-10b in serum was correlated with the level of PLR,providing a new idea for the study of the relationship between inflammation and miRNAs.
