Objective:Through the determination of diabetic rats and diabetic rats before and after myocardial ischemia reperfusion mitochondria Lon protease(Lon protease 1,LonPl)expression level,evaluation of diabetic rats and diabetic rats myocardial ischemia reperfusion myocardial damage and cell oxidative stress levels,explore the diabetes factors on myocardial ischemia reperfusion rats LonPl expression level,the influence of the factors for the study of diabetes increase the specific mechanism of myocardial ischemia-reperfusion injury to provide theoretical referenceMethod:60 healthy and clean grade male SD rats,weighing 200?250g,aged 2-3 months,were randomly divided into 4 groups(n=15):non-diabetic+sham operation group(NS group),non-diabetic+myocardial ischemia-reperfusion group(NI group),diabetic+sham operation group(DS group)and diabetic+myocardial ischemia-reperfusion group(DI group).The DS group and DI group were given streptozotocin 60mg/kg intraperitoneally to prepare the diabetic rat model,and the NS group and the NI group were given sodium citrate/citrate buffer as control.The rat model of myocardial ischemia-reperfusion injury was prepared by NI group and DI group after LAD ligation for 30 min and reperfusion for 120 min.The NS group and DS group were treated with thoracotomy and no ligation.Serum cTnI content was determined by immunohistochemistry,the range of myocardial infarction was determined by TTC staining,LonPl level was determined by Western blot,the activity of mitochondrial aconitase(mt-AC02)was determined by spectrophotometry,and pathological results were observed by HE stainingResult:Scope of myocardial infarction compared with NS group,the DS group and there was no statistically significant difference in serum cTnI levels(P>0.05),and NI and DI group increase myocardial infarction range,elevated serum cTnI levels(P<0.05);Compared with NI group,DI group increase myocardial infarction range,elevated serum cTnI levels(P<0.05);Compared with the DS group,DI group increase myocardial infarction range,higher serum cTnI levels(P<0.05).Compared with NS group,NI group,the DS group and DI group had elevated levels of myocardial LonPl(P<0.05);Compared with NI group,myocardial LonP1 DI group levels(P<0.05);Compared with the DS group,myocardial LonP1 DI group level difference is no statistical significance(P>0.05).Compared with NS group,the DS group myocardial mt-AC02 activity difference has no statistical significance(P>0.05),NI and DI group myocardial mt-AC02 activity(P<0.05);Compared with NI group,DI group myocardial mt-AC02 activity(P<0.05);Compared with the DS group,DI group myocardial mt-AC02 activity(P<0.05).Conclusion:1.Ischemia-reperfusion injury can induce up-regulation of LonPl expression in non-diabetic rats;2.Diabetic factors can also induce the up-regulation of LonPl expression in rat myocardium,but its induction ability is significantly weaker than that of ischemia-reperfusion injury.3.In the state of diabetes,the induction of LonPl in rat myocardium by ischemia-reperfusion injury disappeared,the level of mitochondrial oxidative stress increased,and the myocardial injury was aggravated.
