Objective To investigate the main environmental influencing factors of coronary heart disease,compare and analyze the expression difference of lncRNA ENST00000609755.1 in peripheral blood leukocytes of coronary heart disease patients and control group and its association with environmental factors and coronary heart disease;Construction and preliminary verification of lncRNA ENST00000609755.1 ceRNA regulatory network,providing new research ideas for the epigenetic mechanism of coronary heart disease.Method A hospital-based case-control study was conducted From July 2016 to September 2018,the First Affiliated Hospital of Fujian Medical University and the Union Hospital of Fujian Medical University collected cases and controls in strict accordance with the inclusion criteria and exclusion criteria,and conducted epidemiological investigations and laboratory tests.The expression level of lncRNA ENST00000609755.1 in the case group and the control group was detected by qRT-PCR in large sample population.The combination and interaction of lncRNA ENST00000609755.1 expression level and environmental factors were analyzed by fork analysis.The differentially expressed miRNAs and mRNAs in coronary heart disease were screened by gene chip.The ceRNA regulatory network of lncRNA ENST00000609755.1-miRNA-mRNA was constructed with the core of lncRNA ENST00000609755.1 and qRT-PCR validation of relevant miRNA and mRNA expression levels in human peripheral blood leukocytes.Result(1)Abdominal obesity,family history of coronary heart disease,hyperlipidemia,hypertension,diabetes and lipoprotein a increase the risk of coronary heart disease,and light diet reduces the risk of coronary heart disease(2)qRT-PCR results showed that the expression level of lncRNA ENST00000609755.1 in the peripheral blood leukocytes of the case group was higher than that of the control group.After controlling other risk factors,the high expression of lncRNA ENST00000609755.1 was an independent risk factor for coronary heart disease.Abdominal obesity and lncRNA ENST00000609755.1 expression levels were statistically significant based on the multiplicative interaction(P=0.009).(3)Based ongene chip,lncRNA ENST00000609755.1-miRNA-mRNA ceRNA regulatory network in coronary heart disease was constructed with lncRNA ENST00000609755.1 as a candidate ceRNA.(4)qRT-PCR verification of ceRNA regulatory network in coronary heart disease case group and control group,found that lncRNA ENST00000609755.1 and ELK1 were up-regulated in the coronary heart disease group and the expression level was positively correlated,the expression of miR-150 was down-regulated and negatively correlated with ELK1 expression level.Conclusion(1)Abdominal obesity,family history of coronary heart disease,hyperlipidemia,hypertension,diabetes,and lipoprotein a may be risk factors for coronary heart disease,and light diet may be a protective factor for coronary heart disease..(2)lncRNA ENST00000609755.1 is associated with coronary heart disease.After controlling other environmental risk factors,high expression of lncRNA ENST00000609755.1 may be an independent risk factor for coronary heart disease.Abdominal obesity and lncRNA ENST00000609755.1 expression level have a multiplicative interaction with coronary heart disease,suggesting that coronary heart disease is the result of environmental factors and epigenetic factors.(3)There was a positive co-expression/negative expression relationship between lncRNA ENST00000609755.1-ELK1/miR-150,and it was confirmed that lncRNA ENST00000609755.1 ? miR-150 and ELK1 may have ceRNA regulatory network relationships...
