Objective :To explore susceptibility and interaction of environmental risk factors and vitamin D receptor(VDR)and X-ray repair cross-complementing group 3(XRCC3)single nucleotide polymorphisms of adult acute Leukemia through Case-control study.And provide ideas for etiological research and primary prevention of adult acute leukemia.Methods: A case-control study was conducted in two groups:case group newly diagnosed with acute leukemia treated in Hematology Department of our hospital,non-hematological patients and healthy persons as control group.References were consulted to identify suspicious and environmental risk factors that people were concerned about.All subjects are interviewed with aquestionnaire including history of smoking,X-ray irradiation,house decoration,hair dyeing,history of fever,electromagnetic radiation exposure,living by the road,lack of ventilation,and other environmental risk factors.collect samples of bone marrow or peripheral blood from the two groups.Extracting DNA.Genotyping 5 SNPs of the two genes by PCR technology.General conditions,single environmental factor analysis,genes and genotypes are analysed by X2 test,multiple environmental factors,single nucleotide polymorphism,interaction of environmental risk and SNP are analysed by logistic regression analysis.Results: Smoking history,house decoration,living by the road,and lack of ventilation are statistically significant.Exposure to the factors above may increase susceptibility to adult AL.House decoration is of the highest risk(OR=3.479 95%CI :1.575-7.685),following by lack of ventilation(OR=3.430,95%CI:1.518-7.754),Smoking history(OR=2.679,95%CI:1.175-6.111)? living by the road(OR=2.231,95%CI:1.034-4.813).The frequency of VDR rs7975232 C?A alleles in the case group is 64.70%?35.30%,and 75.00%?25.00% in control group.Distribution of the two alleles in two groups was statistically significant.The frequency of XRCC3 rs861530 T?C alleles in the case group is 58.09%?41.91%,and 46.38%?53.62% in control group.Distribution of the two alleles in two groups was statistically significant.VDR rs7975232 CA and CA+AA genotypes increase the risk of AL,with OR of 2.996 and 2.369,respectively.XRCC3 rs861530 CT and CT+CC genotypes reduced the susceptibility,with OR of 0.159 and 0.306,respectively.Interaction are found between decoration history and VDR rs7975232(OR=2.909,P=0.043),lack of ventilation and XRCC3 rs861530(OR = 0.652;P = 0.046).Conclusion: Smoking history,house decoration,living by the road,and lack of ventilation are risk factors for adult AL.The risk level from high to low is house decoration,lack of ventilation,Smoking history,living by the road.VDR rs7975232 allele A is a susceptible forAL.XRCC3 rs861530 allele Cis a susceptible forAL.VDR(rs7975232)and XRCC3(rs861530)mutations are the risk and protective factors of AL,respectively.There is significant interactions of synergistic effect between house decoration and the VDR rs7975232 mutant genotype,and the interaction between lack of ventilation and XRCC3 rs861530 mutant genotype can reduce the risk of AL.
