BackgroundParkinson's disease(PD)is a common neurodegenerative disorder,the main pathological changes is dopaminergic neurons degeneration and loss in the substantia nigradeath and the survival of dopaminergic neurons arise alpha-synuclein abnormal deposition in the cytoplasm;DJ-1 protein,is a kind of multi-functional protective protein,We found the DJ-1 protein content decreased significantly in midbrain nigra of some sporadic PD patients,The DJ-1 protein has been oxidized and loss its functions.Thus it reveals that the DJ-1 gene is related to familial and sporadic PD.Small RNAs(miRNAs)as a kind of endogenous,stability,and non-coding RNA molecules can regulate target gene expression in transcription level.We hypothesized that DJ-1 protein of the sporadic and familial PD may be regulated by the specific microRNAs.Because of the rich content and stable presence of salivary miRNAs,We detecting the expression of salivary miRNAs to discover its diagnostic value for Parkinson's disease.So far no study of salivary miRNAs in PD patients has been reported at home and abroad.Objective1.RT-qPCR(Reverse transcription–qPCR)technology is used for detecting the expression of salivary miR-874,miR-145-3p in PD patients.the research estimate the value of salivary miRNAs in diagnosis of PD in this respect.Appication of statistical software analysis Statistical differences in the expression of salivary miR-874,miR-145-3p between Parkinson's patients and normal controls.2.The research analysis the correlation between the expression of salivary miR-874,miR-145-3p and age,course of the disease,the severity of condition on PD.MethodsThrough the case-control study,We used the RT-qPCR(Reverse transcription–qPCR)technology to detect the expression of salivary miR-874,miR-145-3p in 30 Parkinson's patients and 30 healthy volunteers.Appication of statistical software analysis Statistical differences in the expression of salivary miR-874,miR-145-3p between Parkinso's patients and healthy volunteers,And the correlation between the expression of salivary miR-874,miR-145-3p and age,course of the disease,the severity of condition on PD.Results1.There are 60 cases of this subjects,the expression of salivary miR-874 was not detected in 32 cases,16 cases from PD group,16 cases from control group.the expression of salivary miR-145-3p was not detected in 25 cases,15 cases from PD group,10 cases from control group.Two kinds of molecules were both positive expression in PD group and the control group,there was no significant difference between them(P values were 1.0,0.19).2.Subgroup analysis:the expression of salivary miR-874 and miR-145-3p are both higher than the control group,which has significant difference between them(P values were 0.041,0.039).The relative expression 2.54 of salivary miR-874 as the boundary between PD group and control group,the sensitivity was 64.3%,and the specificity was 78.6%.The relative expression 2.02 of salivary miR-145-3p as the boundary between PD group and control group,the sensitivity was 64.3%,and the specificity was 78.6%.3.The expression of salivary miRNA-874?miR-145-3p had no clear correlation to age,gender,total RNA concentrations in saliva,UPDRS?,UPDRS?,Hohn-Yahr stage,the sense of smell score,MMSE,MoCA and course of the disease in PD group(P>0.05).Conclusions1.Salivary miRNA-874?miR-145-3p were positive expression in PD group and the control group,there was no significant difference between them(P values were 1.0,0.19).The expression of salivary miR-874,miR-145-3p can not treat as auxiliary diagnosis index.2.The expression of salivary miR-874 and miR-145-3p are both higher than the control group,which has significant difference between them(P values were 0.041,0.039).The relative expression 2.54 of salivary miR-874 as the boundary between PD group and control group,the sensitivity was 64.3%,and the specificity was 78.6%.The relative expression 2.02 of salivary miR-145-3p as the boundary between PD group and control group,the sensitivity was 64.3%,and the specificity was 78.6%.Because of the high specificity,it may be used as a meaningful screening index.3.The expression of salivary miRNA-874?miR-145-3p had no clear correlation to age,gender,total RNA concentrations in saliva,UPDRS?,UPDRS?,Hohn-Yahr stage,the sense of smell score,MMSE,MoCA and course of the disease in PD group(P>0.05).
