In order to study the anesthetic efficacy and safety of sufentanil,and the antagonistic effect of naloxone on sufentanil.This study was designed to research the anesthesia,resuscitation and side effects of different doses of sufentanil on rats and the antagonistic effect of different doses of naloxone on sufentanil.S0,S5,S10,S50,S100 and S200 groups were divided by intravenous injection of normal saline and sufentanil 5,10,50,100 and 200 ?g/kg respectively.Each group among groups of 50,100 and 200?g/kg dose of sufentanil,5min before the injection of sufentanil,were pre-injected with 20,40,80 ?g/kg dose and 40,80,120 ?g/kg dose of naloxone and noted as S50N20,S50N40,S50N80,S100N40,S100N80,S100N120 and S200N40,S200N120,S200N240 naloxone antagonist group.The disappearance and recovery time of ataxia and righting reflex of sufentanil and antagonist group were observed and recorded,the adverse effects were also recorded.The respiratory rate was recorded before and after the injection,and the degree of sedation was scored.The heart rate and mean arterial blood pressure were recorded before and after the injection and blood gas analysis of these groups were performed.The rats of S0 and sufentanil group were subjected to water maze test on the 1st ~ 6th day after injection.The results show that the incidence of disappear of righting reflex and ataxia was 100% among S5 ~ S200 groups,the latency was close to zero.The duration of anesthesia and imbalance of exercise were significantly prolonged with the increasing dose of sufentanil,they were extended from 12.40 min and 36.60 min to 168.10 min and 323.44 min respectively.The degree of adverse reactions gradually increased.One rat died in the S200 group(10%).Respiratory rate,mean arterial pressure,heart rate,oxygen partial pressure and oxygen saturation compared with before administration,the amplitude of decrease of S5 were 28.7%,31.5%,30.1%,45.6% and 34.8%,with the increase of dose of sufentanil,the decrease was increased,the amplitude of decrease of S200 were 32.5%,40.0%,63.5%,58.1% and 55.5%.In the water maze test,there was no significant difference in S5 ~ S200 group compared with S0 group.The naloxone antagonist group of S50N80,S100N120 and S200N240 did not occur the disappearance of righting reflex,the incidence of salivation and convulsions were reduced to zero,heart rate recovery time were respectively reduced by 83.3%,75% and 50% compared with S50,S100 and S200,respiration rate,oxygen partial pressure and oxygen saturation were respectively decreased by 7.8%,0%,15.3%,7.3%,and 9.8%,4.6%,23.2%,8.8%,and 21.6%,3.2%,22.2%,11.5% compared to before administration.This study indicate that single intravenous injection of 5 ~ 200 ?g/kg of sufentanil is 100% anesthesia effective and rapidly onset.Anesthesia effect and adverse effects were doserelated with sufentanil,the rat cardiovascular system and the toxic side effects of respiratory system were dose-related too.Sufentanil had no significant effect on learning and memory function in rats.Pre-injection with naloxone at the doses of 80,120 and 240 ?g/kg completely antagonized the anesthetic effect of sufentanil at the doses of 50,100 and 200 ?g/kg respectively.Antagonism effect of naloxone on sufentanil anesthesia efficacy and adverse effects was dose-related,and can effectively alleviate or even completely reverse side effects of what sufentanil produced on the cardiovascular and respiratory system.
