Colorectal cancer(CRC)is a malignant gastrointestinal tumor.As the disease progresses,the cancer will gradually increase and cause the patient's bowel habits to change,resulting in diarrhea or constipation,accompanied by symptoms such as local abdominal pain or anemia,which will cause the patient to lose weight and reduce immune function.As the incidence and mortality of colorectal cancer continue to increase in China,its threat to the health of our people is gradually deepening.At present,the main clinical treatment options for colorectal cancer have different degrees of side effects on patients.By adjusting the intestinal microbes to improve the treatment results or ensure that patients get a better quality of life in anti-cancer treatment,It is increasingly regarded as an effective and feasible method.In this study,probiotics and effective active ingredients were separated from the traditional fermented food Serofluid dish in the northwest region to study the mechanism of anti-colorectal cancer.In this study,the microorganisms isolated in the Serofluid dish were used to conduct in vitro experiments on colon cancer cells such as HCT116,SW480,and HT29,and their anti-colorectal cancer effects were preliminarily determined by the MTT method.To determine the effect of the active components of Serofluid dish on the cycle and apoptosis of cancer cells,and to explore its anti-colorectal cancer effect through the Transwell migration test,and to determine the important signaling molecules in the PI3 K signaling pathway in mRNA by qRT-PCR and Western Blot Level and expression of corresponding protein levels.The mechanism of anti-colorectal cancer of Serofluid dish cell-free Supernatants SCS-2 was preliminarily explored.Based on the above analysis,the MTT method was used to compare the ability of multiple strains of Serofluid dish bacteria to inhibit colorectal cancer.The results showed that different strains had significantly different inhibitory effects on colorectal cancer.Lactobacillus plantarum YT013 and Lactobacillus paracasei ZJ04 have a significant inhibitory effect on colorectal cancer.The mechanism may be to increasethe expression of E-Cadherin and inhibit the migration ability of colorectal cancer cells.It can inhibit the proliferation of colorectal cancer cells and increase the apoptosis of colorectal cancer cells by regulating the cell cycle and PI3 K signaling pathway.In summary,in this topic,through the separation and purification of Serofluid dish microorganisms,and through the study of the effects of its active ingredients on colorectal cancer cells,the active ingredient SCS-2 of Serofluid dish with inhibitory effect on colorectal cancer was found.
