Gastric cancer is one of the fifth most common tumors in the world,and most patients are currently diagnosed at an advanced stage and have poor prognosis.The aim of this study was to explore the role and molecular mechanism of LINC01272 in the development of gastric cancer.Objective: Using bioinformatics methods,the biological role of LINC01272 in gastric cancer was predicted and to explore the effect of LINC01272 on the migration and invasion of gastric cancer and its molecular mechanism of action.Methods: The original files of the human gastric cancer dataset were downloaded from the TCGA database,and data analysis was performed using R software to select LINC01272 with significant differential expression in gastric cancer as the research object,and the biological function and mechanism of LINC01272 in gastric cancer were predicted by bioinformatics methods,Correlation analysis of LINC01272 with epithelial mesenchymal transition(EMT)related transcription factors was performed,and GO enrichment analysis and KEGG analysis were performed to further predict the signaling pathways affected by LINC01272 in gastric cancer.The expression of LINC01272 in gastric cancer cell lines,gastric cancer tissues and corresponding normal tissues was detected using real-time polymerase chain reaction(RT-PCR).After silencing and overexpression of LINC01272,wound-healing and Transwell assays verified the ability of LINC01272 on the migration and invasion of gastric cancer cells,and RT-PCR and Western-blotting assays examed the expression of EMT-related proteins.Results: The results of bioinformatics analysis showed that LINC01272 was up-regulated in gastric cancer and was associated with gastric cancer stage and lymph node metastasis,and the results of correlation analysis showed that LINC01272 was positively correlated with MMP2 and MMP9,but negatively correlated with TJP1.The results of GO enrichment analysis and KEGG analysis showed that the main signaling pathways affected by LINC01272 were Toll-like receptor signaling pathway,NF-kappa B signaling pathway,and Natural killer cell mediated cytotoxicity.RT-PCR results showed that LINC01272 was significantly highly expressed in gastric cancer cells and tissues.After silencing the expression of LINC01272,the results of wound-healing and Transwell assays showed that LINC01272 inhibited gastric cancer cells migration and invasion,and overexpression of LINC01272 had the opposite effect.Silencing LINC01272,RT-PCR and Western-blotting results showed that MMP-9,N-CAD,ZEB2,and Vimentin expression levels were decreased,while overexpression of LINC01272,the results showed that MMP-9,N-CAD,Twist,and ZEB2 expression levels were up-regulated.Conclusion: In conclusion,our findings suggest that LINC01272 plays an important role in the development of gastric cancer,LINC01272 promoted gastric cancer metastasis ability through regulating the expression of EMT-related proteins and could serve as a potential diagnostic biomarker for gastric cancer.
