Malignant tumor,namely cancer,is the canceration of normal cells in human body under the action of various carcinogenic factors,which makes normal cells become malignant cancer cells,and is a serious threat to human health.At present,the synthetic anti-tumor and anti-oxidation drugs have some limitations in the treatment.The development of natural anti-oxidation and anti-tumor drugs has attracted more and more attention of medical workers and scientific researchers.Fungi contain fungal polysaccharides,which can activate T,B lymphocytes,macrophagesand complement system,enhance the host immune regulation system,so as to achieve the function of anti-oxidation and anti-tumor.At present,the fungi polysaccharides of Hericium erinaceus,Ganoderma lucidum,Shiitake mushroom,Tremella polysaccharide,etc are studied in depth.In this study,the crude polysaccharide PEP of Piptoporus betulinus was extracted by hot water extraction method,and the polysaccharide PEP1 was preliminarily separated and purified by ethanol classification precipitation method.The molecular weight distribution and monosaccharide composition of PEP1 was measured,and the optimum extraction conditions of polysaccharide were studied.The antioxidant and immune activities were also studied in order to provide a scientific theoretical basis for the research and exploration,development and utilization of Piptoporus betulinus.The main results of this study are as follows:1.The yield of polysaccharide is 6.48%,the total sugar content is 65%,the protein content is 12.26%,the glucuronic acid content is 0.24%,the ash content is 1.32%,the monosaccharide composition is mainly glucose(51.86%)and galactose(32.75%),and the molecular weight distribution is not uniform.2.The homogeneous polysaccharide PEP1 is initially isolated and purified.The number average molecular weight is 253652 Da,and the peak shape is symmetrical.It consists of D-Man,D-Glc,and D-Gal,and the content percentages are: 2.23%: 81.97%: 15.80%.3.Response surface methodology is used to optimize the conditions of ultrasonic assist extraction of Polysaccharides from Piptoporus betulinus : water bath time 1 h,water bath temperature 69.16℃,ultrasonic temperature 70℃,ultrasonic time 20 min,ultrasonic power 80.24 W,ratio of liquid to material 30:1,three times of extraction.The yield of polysaccharide is 10.228%.4.The polysaccharide of Betula obliquus can not directly inhibit the proliferation of tumor cells,and can not directly induce apoptosis and necrosis.5、Both PEP and PEP 1 have better ability of scavenging hydroxyl radicals,and the scavenging power of PEP 1 is greater than that of PEP.6.PEP and PEP1 have better ability of scavenging DPPH radicals,and the scavenging power of PEP is greater than that of PEP1.7.PEP and PEP1 can alleviate and inhibit the oxidative damage of A549 and 293 T cells induced by acrylamide,that is to say,the polysaccharide of Piptoporus betulinus has a better antioxidant activity,and PEP1 has a slightly stronger protective effect than PEP.
