Objective To investigate the effect of alcohol extract from a pearl of the head on scopolamine(SCOP)-induced cognitive memory impairment and synaptic plasticity in AD rats and its possible mechanism.Methods 60 SD rats were divided into three groups: normal group,scopolamine group,one pearl alcohol extract treatment group(low dose group,middle dose group,high dose group)and donepezil hydrochloride group(n = 10).After one week of adaptive feeding,one pearl treatment group was given one pearl alcohol extract of the same volume distilled water to stomach for two weeks,and the positive control group was given donepezil hydrochloride Qi,normal group and scopolamine group were infused with distilled water of equal volume.Water maze training was carried out on the 8th day after gavage,and scopolamine 2 mg / kg was injected into all groups except the normal group by intraperitoneal injection of normal saline at the same time.Water maze training was carried out half an hour after injection,and water maze test was carried out on the 14 th day after gavage,i.e.the 7th day after intraperitoneal injection,to observe the spatial learning and memory ability of rats.The activity of cholinergic system enzyme was detected by ELISA and colorimetry,the expression of synapse related protein was detected by Western blot,the injury of Nissl body in CA1 and CA3 areas of hippocampus was detected by Nissl staining,and the brain of each group was observed by Golgi staining Changes of dendritic spine morphology and density in CA1 area of hippocampus.Results 1.The results of water maze experiment showed that the escape latency of scopolamine model group was significantly longer than that of the normal group,and the number of times of crossing the survival platform was significantly reduced,and the escape latency of a pearl drug group on the top of the head could be shortened,which indicated that a pearl alcohol extract on the top of the head could improve the cognitive function of AD model rats;2.The results of ELISA showed that the alcohol extract of a pearl on the top of the head could reduce the expression of AChE activity in the hippocampus of AD model rats;the colorimetric method showed that the alcohol extract of a pearl on the top of the head could increase the activity of ache;3.The Western blotting experiment showed that the medicine group of a pearl on the top of the head could increase synapsin-1,synaptophysin,NMDA and other synaptic proteins and synaptic related kinases in the hippocampus of rats The expression level was dose-dependent;4.Nissl staining showed that the drug group with a pearl alcohol extract from the top of the head could increase the number of Nissl bodies in the hippocampus and promote the repair of neurons in a dose-dependent manner;5.Golgi staining showed that a pearl alcohol extract from the top of the head could increase the density and complexity of dendrite spines in the brain of AD model rats.Conclusion The alcohol extract of a pearl on the head can effectively improve the spatial learning and memory ability of AD model rats induced by scopolamine.The possible mechanism is that it can regulate cholinergic system by inhibiting AChE activity in the brain,increasing AChE activity,improving the level of synaptic protein in the hippocampus of AD model rats,promoting the development of neurons and synapses,and improving dendrites Spine density.
