Objective: to investigate the expression of MT1-MMP in papillary thyroid carcinoma and whether the expression of MT1-MMP is related to clinicopathological characteristics of patients,and to explore the role of MT1-MMP in the progression of papillary thyroid carcinoma.Methods: the surgical specimens of 78 patients who underwent total or partial thyroidectomy from January 2017 to January 2019 in the first hospital of Lanzhou university were collected and made into paraffin sections.Immunohistochemistry was used to determine the positive expression of MT1-MMP in the sections,and the correlation between the positive expression and various clinicopathological features of papillary thyroid carcinoma was analyzed.Subsequently,cell experiments were conducted for further verification.The experiments were divided into the experimental group(with siRNA)and the control group(with PBS).K1 cells were transfected and MT1-MMP gene was silenced.Subsequently,the expression level of MT1-MMP protein and the cell migration capacity were detected by Western blot and cell scratch test,respectively.Results: MT1-MMP has a higher positive rate in papillary thyroid carcinoma than in benign thyroid neoplasms.The expression of MT1-MMP in papillary thyroid carcinoma associated with the presence of lymph node metastasis in patients(P<0.05),but had no significant correlation with gender,age,number of lesions and tumor size(P>0.05).After cell transfection,compared with the control group,the protein level of MT1-MMP in K1 cells of the experimental group was significantly down-regulated,and the cell migration ability was significantly reduced(P>0.05).Conclusion: The positive rate of MT1-MMP in papillary thyroid carcinoma was significantly higher than that in benign thyroid neoplasm.MT1-MMP is highly expressed in papillary thyroid carcinoma.MT1-MMP was not related to the patient's age,gender,tumor size,and number of lesions,but played an important role in the process of lymph node metastasis,promoted the invasion and metastasis of papillary thyroid carcinoma.
