Objectives:1.To observe the changes of ECM in skeletal muscle of sarcopenic rats and explore its possible mechanism.2.To investigate the effects of resveratrol on ECM metabolism,skeletal muscle mass and strength and related mechanisms in aged rats with sarcopenia.Methods: A total of 30 male rats were allocated to 3 groups: the old sarcopenia group(OS group,24 months old),the old resveratrol-treated group(OR group,24 months old),and the young control group(YC group,5 months old).At the end of the 6-week treatment,grip strength,muscle mass,and the protein expression of collagen,MMPs/TIMPs were measured.Results: Compared with the YC group,the OS group had significantly lower relative grip strength,percentage of lean mass,and decreasing expressions of MMPs,with increased expressions of collagen-1,TIMPs protein.Scanning electron microscopy showed that in the OS group,thecollagen fibers were thick and disordered,cross-linking increased.Compared with the OS group,the OR group had higher grip strength and increased MMPs protein expressions,and significantly lower expressions of collagen-1 and TIMPs.SEM revealed that the collagen fibers were finer and arranged more regularly in the OR group than in the OS group.Conclusions: Our study showed that skeletal muscle ECM deposition caused by aging is involved in the development of sarcopenia.Resveratrol regulated the protein expression of MMPs/TIMPs,accelerated ECM metabolism,reduced collagen deposition,and improved skeletal muscle function.
