The Ultraviolet induced skin aging called photoaging.Photoaging skin can represent in a series of cell factors increase such as interleukin, matrix metalloproteinases (MMPs), activated protein (AP-1) and the DNA damage. MMPs can degradable most of the extracellular matrix (ECM), which is the direct cause of photoaging of the skin. Tissue inhibitor of Matrix Metalloproteinase-1(TIMP-1) mRNA is the specificity of the endogenous inhibitor of MMPs,which can inhibit MMPs activation and expression, and then prevent the occurrence of photoaging. Resveratrol is a polyphenol compound which exists in the plants and has many beneficial functions to human body such as anti-angiocardiopathy,Antitumous effect,antioxidant activity,anti-inflammatory,ect. The Development of natural preparations become one of the protective methods of photoaging.Objective①To investigate the effects on the proliferation and the expression of MMPsand TIMPs mRNA of HaCaT Cells after UVB irradiation.②To investigate the effects of Resveratrol on the proliferation and the expression of MMPs and TIMPs mRNA of HaCaT Cells after UVB irradiation. To study the Photo-Protective mechanism of resveratrol from UVB-induced damage on HaCaT cells.Methods①Draw cell growth curve;②The cultured HaCaT cells were irradiated with UVB of30、60、90mJ/cn2respectively, The survival rate was measured by MTT assay;③The MMP-1、MMP-9、TIMP-1mRNA expression were measured by RT-PCR after24h of different dosages UVB irradiation.④HaCaT cells were cultured with different concentration of resveratrol and the proliferation of HaCaT cells was detected with MTT assay.⑤HaCaT cells were cultured with0.1and0.5mmol/L resveratrol after30mJ/cm2(90mJ/cm2)UVB irradiation,and the expression of MMP-9and TIMP-1mRNA (MMP-1mRNA) of HaCaT cells were measured by RT-PCR.Results①The survival rate increased gradually in normal HaCaT cells as time extends. At24h after UVB irradiation, the survival rate of HaCaT cells was lower than that in control group at the same time point (p<0.05).②The expression of MMP-1、MMP-9mRNA were lower in control group. The MMP-9mRNA expression was significantly increased at24h after irradiation of three different dosage of UVB compared to that in the normal control group at the same time point(p<0.05). Comparative differences between groups were statistically significant (P<0.05).MMP-1mRNA expression was significantly increased at the24h after90mJ/cm2UVB irradiation (p<0.05). The expression of TIMP-1mRNA was decreased of three different dosage of UVB irradiation compared to that in the normal control group (P<0.05).③The proliferation of HaCaT cells were not obviously affected while cultured with different concentration of Res (p>0.05).④After24h of cultured with0.1mmol/L Res, the MMP-1and MMP-9mRNA expression were not decreased and TIMP-1was not increased compared with the control group (P>0.05). After24h of cultured with0.5mmol/L Res, the MMP-1and MMP-9mRNA expression were decreased and TIMP-1was increased compared with the control group (P<0.05).Conclusion①UVB irradiation can inhibit the proliferation of HaCaT cells. It also can induce the expression of MMP-1、MMP-9,and decrease the expression of TIMP-1on HaCaT cells which makes the MMP/TIMP ratio imbalanced. This maybe have some relationship in the pathogenesis of skin photoaging.②Resveratrol can signally inhibit the change of MMP-1、MMP-9and TIMP-1mRNA expression after UVB irradiation in HaCaT cells and may have certain protective effect on skin photoaging. It maybe play a repairment role in UVB-induced skin damage.
