Alzheimer 's disease(AD)is a multifactorial disease.Epigenetic abnormalities,genomic changes,environmental risk factors,and cerebrovascular damage all cause premature and progressive neuronal loss.According to statistics from the medical community,about 2.5% to 5% of the elderly over 65 years old will suffer from the disease.In AD cases,amyloid plaques(SP)formed by A? and neurofibrillary tangles(NFTs)formed by Tau proteins are two obvious neuropathological features of brain tissue of AD patients.With the development of economy and the improvement of living standards,the dietary fiber level in residents' diet structure is too low.In order to improve the increasingly unbalanced diet structure,cereal drinks will be more and more concerned in the future.Grain beverages are nutritious foods processed with corn,barley,wheat,barley,corn and other cereal foods as the main raw materials.They aim to improve the health of our people by changing the diet structure.In addition,studies have found that diabetes,heart disease,stroke and other diseases can be alleviated by the intake of whole grains,reducing the incidence of disease.Cereal drinks have many other functions to promote human health.A high-fiber diet can reduce the incidence of colon cancer.This thesis mainly studies the role of a cereal fermented drink,zymolytic grain extract(ZGE)in reducing the cytotoxicity of A?42 oligomers.The total supernatant of ZGE(ZGETS)is separated into three sections:high molecular weight,middle and low molecular weight(ZGEHM,ZGEMM,ZGELM).This study found that ZGETS and ZGELM have a significant inhibitory effect on the cytotoxicity of A?42 oligomers,indicating that this effect is exerted by the small molecule meterials in ZGETS.We selected three other cells: human embryonic kidney cells(HEK 293),human cervical cancer cells(HeLa),and hepatoblastoma cells(HepG 2)as controls to SH-SY5 Y,and also confirmed this effect of ZGE.ZGE acts on SH-SY5 Y cells,causing nerve cell protrusions to become longer,and the number of cells also increases to a certain extent,showing a certain trophic effect on cells.These effects are not observed in the other three cells.Through the aggregation experiment of A?42,it is concluded that ZGTLM has a certain inhibitory effect on the aggregation of oligomers of A?42.Analysis of amino acids,proteins and carbohydrates in ZGE confirmed that the main component of ZGELM is carbohydrates.The types of sugar in ZGE were analyzed and identified,and it was determined that there were mainly fructose,glucose and some oligosaccharides in ZGELM.According to the specific analysis of the four states of A?42 by each part of ZGE,ZGELM has certain specificity for the four states of A?42,among which the oligomer has the strongest specificity.,and after analyzing the survival rate of cells and observing cell morphology by using carbohydrates identified in ZGE and A?42 oligomers act together on SH-SY5 Y,it was finally determined that the substances that inhibit the cytotoxicity of A?42 oligomers should be oligosaccharides in ZGE.Due to the complexity of oligosaccharides in ZGE,the substances that play a major neuroprotective role need to be further analyzed in detail.
