Isoform-Specific Expression Of 14-3-3mRNA In Human Cervical Cancer And Cervical Intraepithelial Neoplasia

Objective: To study the expression of 14-3-3 mRNA in the chronic cervicitis, cervical intraepithelial neoplasia and cervical cancer.Methods: The expression of 14-3-3 mRNA was measured by RT-PCR in the chronic cervicitis, cervical intraepithelial neoplasia and cervical cancer.The experimental datas were obtained by ImageTool software. Statistical analysis was performed using SPSS for windows 11.5 software.Results: The level of every isoform of 14-3-3mRNA in the chronic cervicitis(control),CINⅠ,CINⅡ,CINⅢand cervical cancer(CC) were listed as follow: (1)14-3-3ζmRNA:The expression of 14-3-3ζmRNA in the control group, CINⅠgroup,CINⅡgroup,CINⅢgroup and CC group was 0.68±0.01, 0.71±0.05,0.73±0.07, 0.67±0.06 and 0.69±0.07 respectively, which didn't indicate a significant differences (P>0.05); (2)14-3-3εmRNA:The expression of 14-3-3εmRNA in the control group, CINⅠgroup, CINⅡgroup, CINⅢgroup and CC group was 1.22±0.09,1.28±0.10,1.29±0.07,1.27±0.12 and 1.21±0.15 respectively, which didn't indicate a significant differences(P>0.05); (3)14-3-3θmRNA: 14-3-3θmRNA was lower transcribed significantly in CC group(0) than that in other groups(p<0.01); there were no significant difference between in every CIN group and in control group(p>0.05); and there was no significant difference among CIN group(p>0.05);(4)14-3-3σmRNA : The expression of 14-3-3σmRNA in the control group, CINⅠgroup, CINⅡgroup,CINⅢgroup and CC group was 0.75±0.02,0.66±0.01, 0.60±0.04,0.35±0.02 and0.28±0.05 respectively, the expression of 14-3-3σmRNA decreased from control group to CC group(p<0.05); (5)14-3-3γmRNA:The expression of 14-3-3γmRNA in the control group, CINⅠgroup, CINⅡgroup,CINⅢgroup and CC group was 1.50±0.25,1.36±0.11,1.48±0.12,1.37±0.24 and 1.58±0.29 respectively, which didn't indicate a significant differences(P>0.05); ( 6 )14-3-3ηmRNA : The expression of 14-3-3ηmRNA in the control group, CINⅠgroup, CINⅡgroup,CINⅢgroup and CC group was 0.75±0.06,0.80±0.07,0.81±0.02,0.77±0.03 and 0.74±0.06 respectively, which didn't indicate a significant differences(P>0.05);(7) 14-3-3βmRNA:There was significant difference when control group compared with the CINⅡgroup,CINⅢgroup or CC group(0.74±0.06) (P<0.01). There was also significant difference between every CIN group and CC group(p<0.05).the expression of 14-3-3βmRNA increased from CINⅠgroup to CINⅢgroup(0.77±0.03)(p<0.05).Compared control group with CINⅠgroup (0.80±0.07), there was no significant difference (P>0.05).Conclusion:1. 14-3-3 mRNA showed a basic expression in chronic cervicitis.2. The expression of 14-3-3σmRNA decreased with the progress of cervical lesion.3. The expression of 14-3-3βmRNA was up-regulation in CINⅡ,CINⅢand cervical cancer.4.14-3-3θmRNA was unexpressive in the cervical cancer.5.The expression of 14-3-3σ,βandθmRNA was abnormal in the cervical pathological changes ,which suggested these three isoforms may be involved in the development of cervical pathological changes.
