Objective:In this study,exosomes in circulating blood were extracted and their particle size and concentration were determined.Then qPCR was used to detect the expression of miR-21 and miR-92 a in serum exosomes in NPC patients,and to explore serum exosomes.The diagnosis and clinical significance of the expression of miR-21 and miR-92 a in NPC.Methods:Peripheral blood samples were collected: 37 patients with NPC and 29 healthy controls;miRNAs in serum exosomes were extracted;serum exosomes were isolated and extracted by ultrafiltration,and the exosome was extracted by TNA method for particle size and concentration.Determination.The expression of miR-21 and miR-92 a in serum exosomes was detected by qPCR.The expression levels of quantitative miRNAs were calculated.The measurement data were expressed as(?)ąs,and the mean comparison between groups was analyzed by one-way variance method.T test was used to analyze the correlation between clinicopathological parameters of NPC patients and the expression levels of miR-21 and miR-92 a in serum exosomes.Paired T test was used to compare the expression level of miR-92 a in miR-21 before and post-chemotherapy,post-radiation.All the statistically analysis were performed using SPSS23.0 and P values less than 0.05 were considered to be statistically significant.Results:1.The microvesicles extracted using the ultra-fast filtration method ranged mainly in the range of 40-150 nm with a peak of 86 nm and a concentration of about 4.34 e + 008 particles/ml.2.The expression level of serum exosome miR-21 in NPC patients is significantly higher than that in healthy controls.3.The expression level of serum exosome miR-92 a in NPC patients was significantly lower than that in healthy controls.4.The expression levels of serum exosomes miR-21 and miR-92 a in NPC patients are related to T stage and N stage.5.The expression level of serum exosome miR-21 in NPC patients was correlated with the end of radiotherapy;the expression level of miR-92 a before treatment was correlated with post-chemotherapy and post-radiation treatment.Conclusion:miR-21 and miR-92 a in serum exosomes are closely related to the occurrence and development of nasopharyngeal carcinoma.They may be potential new biomarkers for the diagnosis and prognosis of nasopharyngeal carcinoma,and monitoring their dynamic changes may be helpful for early detection.Recurrence and metastasis.
