Objective:Immunohistochemistry(IHC)was used to detect the expression of phosphorylation signal transduction and activator of transcription 3(p-STAT3)in breast invasive ductal carcinoma and adjacent normal breast tissues.To analyze the expression level of p-STAT3 in breast invasive ductal carcinoma and its correlation with clinicopathological characters,body mass index(BMI)and breast density.Combined with patients' long-term living conditions to explore the predictive value and significance of p-STAT3 in clinical prognosis of patients with breast invasive ductal carcinoma.Method:A retrospective analysis of 82 cases of invasive breast cancer patients who underwent surgical treatment in the First Affiliated Hospital of Jinan University from January 2013 to March 2016,another 48 specimens from normal adjacent tissues served as controls,and follow-up of postoperative survival status in these patients with breast cancer.Immunohistochemical was used to detect the expression of p-STAT3 in paraffin sections of 82 cases of breast invasive ductal carcinoma and 48 cases of adjacent normal breast tissues.Chi-square test was used to analyze the difference of p-STAT3 expression in normal tissues and breast cancer tissues,the correlation between p-STAT3 expression and clinical pathological features,BMI and breast density in breast invasive ductal carcinoma.Survival analysis was performed using the Log-Rank test to explore the predictive value of p-STAT3 for clinical prognosis in patients with breast invasive ductal carcinoma.Results:1.Among the 82 patients with invasive ductal carcinoma of the breast,the average age was 49.6 years,the youngest was 37 years old,and the largest was 76 years old.2.Among the 82 breast cancer specimens,45 patients were positive for p-STAT3,37 patients were negative for p-STAT3,and the positive rate was 54.9%.Among the 48 adjacent normal tissues,11 were p-STAT3 positive,37 were p-STAT3 negative,and the positive rate was 22.9%;the difference between the two was statistically significant(P=0.00<0.05).3.By analyzing ER status,PR status,HER-2 status,status of lymph node metastasis,TNM stage,menstrual status,breast density,BMI and other pathological features.It was found that p-STAT3 was closely related to ER positive,PR positive,smaller tumor diameter,earlier TNM stage and molecular typing,but there was no significant correlation with HER-2 status,Ki-67 value,histological grade,lymph node metastasis,BMI and breast density.4.Log-Rank survival analysis indicated that the disease-free survival(DFS)of patients with positive p-STAT3 was longer,P=0.028<0.05.Conclusion:1.Compared with normal breast tissue,the proportion of p-STAT3 positive in breast invasive ductal carcinoma was higher.2.breast cancer cells with positive expression of p-STAT3 were less malignant and less invasive.3.p-STAT3 can be used as a biomarker for predicting the prognosis of patients with breast invasive ductal carcinoma,and has potential clinical application value.
