Yupingfeng polysaccharide is made from the medical herba compound of astragalus,atractylodes,divaricatesaposhnikovia root,which are extracted through poaching,alcohol precipitation and spray drying.In this study,Specific Pathogen Free?SPF?male mice were used as the research object to construct a non-alcoholic fatty liver disease?NAFLD?model to probe the treatment effect of Yupingfeng polysaccharide.Firstly,the mice were randomly divided into two groups,normal control group?group C?and fatty liver group?group F?.Mice in group F were received intraperitoneal injection with 10%CCl4 solution,while group C mice were injected with normal saline.After four weeks,the mice of group F were randomly divided into low-dose group?group L?,medium-dose group?group M?,high-dose group?group H?and natural recovery group?group N?.In group L,M and H,mice were respectively given 1667 mg/L,2334 mg/L and 4668 mg/L Yupingfeng polysaccharide0.5 ml once a day.and group C and N were infused with normal saline.Blood was sampled from mice eyes,and the the serum contents of total cholesterol?TG?,total bile acid?TAB?,triglyceride?TCHO?,alanine aminotransferase?GPT?and glutamic oxaloacetic aminotransferase?GOT?in serum of mice were measured.The results showed that the contents of five indexes in serum of mice in groups L,M and H were lower than those in group N.The content of TCHO in group H was significantly lower than those in group N?P<0.05?,and the content of TBA and GOT was significantly higher than those in group C?P<0.05?.The content of TBA and GPT content in group M was significantly lower than those in group N?P<0.05?,and the content of TCHO was significantly higher than those in group C?P<0.05?.The content of TCHO,TBA and TG content in group L was significantly higher than those in group C?P<0.05?.The relative expression levels of genes CYP7A-1,FGF-15,ASBT and FXR were determined by fluorescence quantitative method.The results showed that the relative expression of genes ASBT and FXR in mice of group N was significantly higher than those in group C?P<0.05?.The relative expression of gene FGF-15 in group M was significantly lower than those in group N?P<0.05?.The relative expression of gene CYP7A-1 in group L was significantly higher than those in group N?P<0.05?,while the expression of gene FGF-15 was significantly lower than those in group N?P<0.05?.By 16s rDNA sequencing and data analysis,significant differences in intestinal flora between group C and N,L,M and H mice were detected?P<0.05?.There were also significant differences in intestinal flora between group N and H mice?P<0.05?.At Bacteriophyta level,the first dominant flora in intestinal tract of mice in group C was the Bacteroidetes,and the Bacteroidetes was the second group.On the contrary,the Firmicutes was the first dominant group in groups L,N and H,and the second dominant group was the Bacteroidetes.Similarity,Firmicutes was the first dominant group in group M,but the Bacteroidetes accounted for less.At bacterial generic level,the content of Bacteroides and Murbaculum in group C were significantly more than those in group N,M and L?P<0.05?;Bacteroides in group L was significantly more than those in group N?P<0.05?;Murbaculum in group H was significantly more than those in group N,L and M?<0.05?.In conclusion,NAFLD would cause hepatocyte damage,excessive increase of serum biochemical indicators,abnormal relative expression of metabolic genes and decrease the number of the dominant intestinal flora in mice.Yupingfeng polysaccharide could regulate the metabolism of intestine and liver through the mechanism of intestinal-hepatic axis to promote the recovery of liver function and steady the structure of intestinal flora,which contributes to the treatment of NAFLD in mice.The results from this study provides a new method and idea for the therapy of NAFLD in animal husbandry.
