Objective To establish the animal model of asthmatic rats,and to explore the mechanism of compound allergic Decoction in the prevention and treatment of asthma by observing the effect of compound allergic Decoction on the thickness of airway wall,the deposition of type I and III collagen and the expression of TGF-?1 in airway wall of asthmatic rats.Methods 1.Modeling: the establishment of asthma model is divided into two parts: basic sensitization and local stimulation.Rats were injected with 10% ova suspension 1ml(including ova 100 mg + aluminum hydroxide 100 mg + 0.9% normal saline 1ml)into the right lower abdominal cavity on the first and eighth days of the experiment to complete basic sensitization.From the 15 th day of the experiment,rats in each group were placed in an incompletely sealed transparent plastic atomizing box of the same size,empty In the model group and each treatment group,1%,1.5%,2%,2.5% and 3% ova solution were used to induce asthma in the first,second,third,fourth and fifth weeks.2.Experimental process: 60 Wistar rats were randomly divided into 6 groups according to 10 rats in each group: blank group(A),asthma model group(B),western medicine dexamethasone group(C),traditional Chinese medicine compound anaphylaxis Decoction low dose group(D),compound anaphylaxis Decoction medium dose group(E),compound anaphylaxis Decoction high dose group(F).On the 15 th day of the experiment,rats in each group were given gavage before each atomization,group A and group B were given gavage with distilled water,group C was given gavage with dexamethasone,group D was given gavage with low-dose compound anaphylaxis decoction,group E was given gavage with medium dose compound anaphylaxis decoction,group F was given gavage with high-dose compound anaphylaxis Decoction once a day for 35 days.The physical signs of the rats were observed within 30 minutes after each gavage operation,and the effects of the drugs on the rats in each group were recorded.3.Detection index: 24 hours after the completion of intragastric administration on the 35 th day of the experiment,anesthetized rats from group A to group F,then collected samples of their trachea and lung tissues,completed the relevant operations of hematoxylin eosin(HE)staining,detected and compared the airway wall thickness of rats in each group,and measured and compared the expression of collagen I,collagen III and TGF-?1 by immunohistochemistry.Results: 1.General situation of rats: rats in group A did not have asthma;rats in group B had representative asthma symptoms,such as shortness of breath,irritability,etc.;rats in group C,D,E and F all had asthma symptoms,but improved compared with group A,the two groups with the best improvement effect were group C and group E.2.HE staining and measurement results of lung tissue in rats: there was no special change in group A;eosinophils were found around airway and blood vessels in group B,EOS)was found,and the airway wall was significantly thickened compared with group A,the difference was statistically significant(P < 0.05);in group C,D,e and F,the airway wall thickness was significantly smaller than that in the model group(P < 0.05),which was statistically significant.Therefore,it was concluded that the traditional Chinese medicine Compound Guo Min Jian and dexamethasone could reduce the airway wall thickening in asthmatic rats.3.The contents of collagen I,collagen III and TGF-?1 were measured by immunohistochemistry the content of collagen ?,? and TGF-?1 in group A was significantly higher than that in group B(P<0.05).The content of collagen ? and TGF-?1 in group C,D,E and F was much lower than that in group B(P<0.05).Dexamethasone and dexamethasone can reduce the content of collagen ? and TGF-?1 in asthmatic rats,but have little effect on collagen ?.Conclusions: 1.Compound anaphylaxis decoction can inhibit the thickening of airway wall,the deposition of type ? collagen and the expression of TGF-?1 in the process of airway remodeling,so as to inhibit the airway remodeling of asthma.2.In terms of inhibiting the thickening of airway wall,the deposition of type ? collagen and the expression of TGF-?1,the middle dose compound allergic decoction has the same effect as the Western drug dexamethasone,and the adverse reactions are smaller than dexamethasone.3.There was a positive correlation between TGF-?1 and type ? collagen in the process of airway remodeling in asthma.
