| Objective: To evaluate the effect of Transforming growth factor-β1 and Interferon-γon the expression of Connective tissue growth factor in the airway epithelial cell and the signaling pathway of this course.Methods: Primary airway epithelial cell were obtained from rabbit trachea and cultured in vitro. After cell grew to confluence and were devided into groups. (1) The normal cultured airway epithelial cell were divided randomly into 4 groups: TGF-β1 6h,12h,24h,36h group. This groups were stimulated by TGF-β1(10ng/ml) for 6,12,24,36 hours respectively. The expression of CTGF proteins were measured by immunocyte- chemistry; (2) The normal cultured airway epithelial cell were divided randomly into 4 groups: the normal control group and TGF-β1 1ng/ml,5ng/ml,10ng/ml group. The expression of CTGF proteins were measured by immunocyte- chemistry, the expression of Smad7mRNA were measured by in site hybridization; (3) The normal cultured airway epithelial cell were divided randomly into 4 groups: TGF-β110ng/ml with IFN-γ0 ng/ml,10ng/ml,20ng/ml,50ng/ml group. The expression of CTGF proteins were measured by immunocyte- chemistry, the expression of Smad7mRNA were measured by in site hybridization.Result:(1) The time-effect relationship of TGF-β1 and the expression of CTGF: The expression of CTGF proteins in TGF-β1 6h group(0.2518±0.04402),12h group(0.4547±0.01835),24h group(0.6533±0.0652),36h group(0.5513±0.0391)were significantly higher than that in the normal control group, P<0.01; (2) The dose-effect relationship of TGF-β1 and the expression of CTGF: The expression of CTGF proteins in TGF-β1 5ng/ml group(0.4038±0.0293),10ng/ml group(0.6533±0.0127)were significantly higher than that in the normal control group(0.1097±0.0224), P<0.01, and there were significant difference in TGF-β1 1ng/ml,5ng/ml,10ng/ml group in term of CTGF proteins expression , P<0.01; (3)The effect of TGF-β1 on the expression of Smad7mRNA: The expression of Smad7mRNA in TGF-β1 1ng/ml group(0.1528±0.0641)5ng/ml group(0.193±0.017),10ng/ml group(0.206±0.0198)were not significantly higher than that in the normal control group(0.1519±0.0582), P>0.05; (4) The effect of IFN-γon the expression of CTGF proteins: The expression of CTGF proteins in IFN-γ10ng/ml group(0.5005±0.0243),20ng/ml group(0.3466±0.0224),50ng/ml group (0.1417±0.0243)were significantly lower than that in the TGF-β110ng/ml group, P<0.01; (5)... |
