Objective: In this study,the relative expression of DACT2 and PITX2 m RNA in normal control group,colon adenoma group,and colon cancer membrane group was jointly detected,and the interaction between the two DACT2 and PITX2 the malignant transformation of colon adenoma tissue was initially explored.And it may play a role in providing a certain idea for improving the early diagnosis rate of colon cancer and providing more possibilities for more effective treatment of colon cancer patients.Methods: 1.Experimental grouping: 90 specimens were divided into 3 groups,30 cases in control group,30 cases in colon adenoma group and 30 cases in colon cancer membrane group.2.Source of cases: Patients who were admitted to the Department of Gastroenterology of the Third Affiliated Hospital of Inner Mongolia Medical University from December 2018 to October 2019 were included in the patients who underwent colonoscopy and obtained biopsy specimens under the microscope.3.Experimental method: Real time q PCR method was used to detect the expression of DACT2 m RNA and PITX2 m RNA in normal control group,colon adenoma group and colon cancer membrane group.Results: 1.The relative expression levels of DACT2 m RNA detected in the control group,colon adenoma group and colon cancer membrane group were 2.983 ±0.999,1.941 ± 1.001,0.922 ± 0.342,and the relative expression levels in the three groups gradually decreased,the highest content in the normal control group.The difference between the normal control group and the colon adenoma group and the colon cancer membrane group was statistically significant(P<0.001).The comparison between the colon adenoma group and the colon cancer group was very significant(P<0.001).2.The relative expression levels of PITX2 m RNA in the above three groups were 0.915 ± 0.256,1.390 ± 0.532,1.704 ± 0.49,which gradually increased in the three groups,and the content of the control group was the lowest.Compared with the colon adenoma group and colon cancer group,the difference was statistically significant(P<0.001),the relative expression content of colon adenocarcinoma group was higher than that of colon adenoma group.There was a significant statistical difference between the two(P<0.001).3.The expressions of DACT2 and PITX2 m RNA in the three groups are related.In the normal control group,the expression of DACT2 and PITX2 m RNA is negatively correlated,and the difference is statistically significant(r =-0.483,P<0.05);in colon adenomas Group,DACT2 was significantly negatively correlated with PITX2 m RNA expression(r =-0.403,P<0.001);in colon cancer group,DACT2 was significantly negatively correlated with PITX2 m RNA expression(r =-0.733,P<0.001).Conclusions: 1.The expression of DACT2 m RNA in the three groups is significantly different,showing a gradual decline trend,and the lowest expression level in the colon cancer group.It is speculated that the DACT2 gene abnormal expression has a certain correlation with the occurrence and progression of coloncancer,and may play a role Role,suggesting that the expression level of DACT2 can be used as a reference marker to improve the diagnosis rate of early colon cancer to a certain extent.2.The expression levels of PITX2 m RNA were different in the three groups,with the highest expression level in the colon cancer group,followed by the colon adenoma group,and the overall trend gradually increased,suggesting that the gene may play a positive role in the process of colonic malignant transformation effect.Therefore,we can further inhibit the occurrence and progression of colon cancer through various techniques to hinder the expression of this gene,which is not only helpful for improving the accuracy of early diagnosis of colon malignant tumors,but also of great significance for providing further treatment to patients.3.The expression of DACT2 and PITX2 was negatively correlated in the three groups,suggesting that the two may play a role in the process of normal colon tissue-colon adenoma-colon cancer,and the interaction between the two.Then the combined detection of DACT2 and PITX2 expression levels may open up new ideas for improving the early diagnosis rate of colon cancer and subsequent treatment.
