Objective: The purpose of this study was to jointly detect the expression levels of DLC-1 and FAK in normal colon mucosa,colon adenoma and colon cancer tissue,and to explore the relationship between them and the clinical significance of colonic mucosa carcinogenesis.To provide a marker for the early diagnosis of colonic mucosal malignancy and a theoretical basis for the early treatment of colonic malignant tumors.Methods:1 Groups: This study contained three groups,and there were 30 cases in the normal control group,30 cases in the colon adenoma group,and 30 cases in the colon cancer group.2 Specimen collection: The collection time range was from December 2017 to October2018;All specimens were collected from the Department of Gastroenterology outpatients or inpatients at the Third Affiliated Hospital of Inner Mongolia Medical University;Colonic mucosa specimens obtained during colonoscopy or treatment were divided into three groups according to pathological diagnosis: normal colon,colon adenoma and colon cancer.3Experimental methods: Real-time fluorescence quantitative PCR was used to detect the expression of DLC-1 and FAK in normal colon mucosa,colon adenoma and colon cancer tissues.Result: 1 The relative expression levels of DLC-1 mRNA in normal colon mucosa,colon adenoma and colon cancer tissue were 0.889±0.090 ? 0.614±0.071 ? 0.216±0.059,respectively.There were very significant differences(P<0.001),the expression was gradually decreasing trend.The relative expression levels of FAK mRNA in normal colon mucosa,colon adenoma,and colon cancer tissues were 0.674±0.219 ? 1.329±0.375 ? 3.136±0.454,respectively.There were very significant differences(P<0.001),the expression was gradually increasing trend.3 In the normal control group,there was a negative correlation between the expression of DLC-1 and FAK mRNA and the difference was statistically significant(r=-0.394,P<0.05);In colonic adenoma group,there was a negative correlation between theexpression of DLC-1 and FAK mRNA and the difference was statistically significant(r=-0.672,P<0.001);In colon cancer group,there was a negative correlation between the expression of DLC-1 and FAK mRNA and the difference was statistically significant(r=-0.812,P<0.001).Conclusion: 1 The expression of DLC-1 in normal group,adenoma group and cancer group decreased gradually,which indicated that the abnormal expression of this gene is associated with the carcinogenesis of colon adenoma,which may play a certain role in tumor suppression,suggesting that the expression level of DLC-1 can reflect the risk of carcinogenesis of colon adenoma to some extent,which can be used as one of the main reference indicators for early diagnosis of colon cancer.2 The expression of FAK gene in normal group,adenoma group and cancer group increased gradually,which suggested that overexpression of this gene might promote the carcinogenesis of colon adenoma.Therefore,we can further hinder or delay the development of colon adenoma to colon cancer by interfering with the expression of FAK,which were of great significance for the early diagnosis and treatment of colon cancer.3 DLC-1 and FAK played a role in the carcinogenesis of colon adenoma,and the relationship between them was negatively correlated,suggested that DLC-1 might directly or indirectly down-regulate the expression of FAK,which together affected the carcinogenesis of colon adenoma.Joint detection of DLC-1and FAK would contribute to the early diagnosis of colon cancer and provide a new direction and ideas for the treatment of colon cancer.
