Objective: Hepatocellular carcinoma(HCC)is one of the most common malignant tumors with high morbidity and mortality.China is one of the countries with the heaviest burden of HCC,with nearly 50% of new cases and deaths worldwide.A large number of studies have shown that exosome mi RNAs are involved in the growth,metastasis and angiogenesis of HCC,and play an important role in the diagnosis,treatment and prognosis of HCC.It has been reported that the abnormal high expression of mi R-155 in HCC is related to tumor progression.However,the expression and clinical significance of mi R-155 in serum exosomes have not been reported.In this study,serum exosome mi R-155 was used as the research object to detect its expression level in patients with HCC and healthy controls,and to analyze the relationship between the expression level and clinical pathological indexes,to explore the diagnostic value of serum exosome mi R-155 in HCC,and to provide theoretical basis for its further clinical application.Methods: The serum exosomes were purified and identified from 33 patients with hepatocellular carcinoma and 15 healthy controls which were collected from Hebei General hospital.Then mi RNA was extracted,and the expression level of mi R-155 was detected by RT-PCR.The relationship between mi R-155 expression and the clinical pathological parameters,such as age,sex,HBs Ag,AFP level,cirrhosis,tumor size and TNM stage,was further analyzed.Results: The expression of exosomal mi R-155 in serum of patients with liver cancer was significantly higher than that of healthy controls,and the relative expression level was 6.3 times higher than that of healthy controls.The high expression of exosomal mi R-155 was significantly correlated with tumor size,TNM stage and the AFP level(P<0.05),but not with age,gender,HBs Ag and liver cirrhosis(P>0.05).Conclusion:Increase of serum exosomal mi R-155 levels in patients with hepatocellular carcinoma is related to the cancer development.The detection of mi R-155 may be helpful for the clinical diagnosis of hepatocellular carcinoma.
