| Objective: In order to prove the scientific basis of the prescription for long-term clinical experience of the tutor,this paper studied the mechanism of action of tonifying kidney,nourishing and activating blood Chinese medicine on follicle-stimulating development and treatment of ovarian diseases from three aspects of theoretical overview,animal experiments and clinical case analysis.Methods: firstly,the etiology and pathogenesis of follicular development in Chinese and western medicine were analyzed.Secondly,through animal experiments,female SD rats were randomly divided into 7 groups,and 8 distilled water(1ml/100g)were randomly selected for the blank control group,and the remaining 6 groups were given triptolide(400g/kg,60d)as the exploration dose of the previous research.After successful modeling,it was divided into blank group,Triptolide group,western medicine group,high and low dose group of traditional Chinese medicine,NVP-BEZ235+ high dose group of traditional Chinese medicine.Each dose was administered according to the experimental setting.After continuous gavage for 2weeks,blood samples were collected to detect blood hormone levels and progress the hemorheological analysis.After blood collection,the rats were sacrificed,their ovaries were taken out and weighed,and then,their ovarian histology was observed.Extract proteins and genes,and the expression levels of PI3 K,Akt,m TOR,Lamp2,cathepsin-d and genes were detected by flow cytometry and polymerase chain reaction.Finally,3 cases of complete clinical trials in which the mentor used XJCRTSZW in the treatment of follicular dysplastic diseases were selected,and the efficacy was analyzed.Results :(1)compared with the normal control group,the rats in the Triptolide model group had estrous cycle disorder,increased number of atret follicles,elevated the levels of FSH?LH and AMH,decreased ovarian wet weight,ovarian index,and E2 levels,and the number of follicles at all levels,corpus luteum,and granular cell layer thickness,with statistically significant differences(p < 0.01).(2)compared with Triptolide group,western medicine,high and low dosetraditional Chinese medicine group ovaries index,wet weight and E2 level were increased,the number of atresia follicles and the levels of FSH?LH?AMH weredecreased,all levels of ovarian follicle,corpus luteum number,granular cell layer thickness were increased,the difference had statistical significance(p < 0.01),and the effect of western medicine group and high dose group of Chinese medicine.(3)compared with Triptolide group,the expression of autophagy related molecular LAMP2?Cathepsin D in the western medicine and high and low dose of traditional Chinese medicine group were deccreased,and m RNA level and protein level are the same,statistically significant difference(p < 0.01),and the signal path shaft related protein phosphorylation of PI3 K,AKT and m TOR levels increased significantly(p < 0.01),but had no significant effect on its m RNA,and after joining autophagy inducer,the influence of high dose group on above molecules disappear,statistically significant difference(p < 0.05 or p < 0.01).Conclusion :(1)after the model construction of Triptolide,the rats presented estrous cycle disorder,hormone and ovarian histology were consistent with the manifestations of ovarian function injury,indicating that Triptolide could successfully establish the model of follicular development disorder.(2)XJCRTSZW can improve the serum hormone level of rats,increase the ovarian index and ovarian wet weight,restore the number of follicles and luteum at all levels,and increase the thickness of granulosa cell layer.(3)XJCRTSZW can significantly reduce the whole blood viscosity,red cell assembling index and erythrocyte sedimentation rate,and play the role of traditional Chinese medicine compound in promoting blood circulation and nourishing blood on the basis of kidney tonifying(4)the damage of rat ovarian function caused by Triptolide may be induced by autophagy by control the PI3K/AKT/m TOR pathway.Tonifying kidney,activating blood and nourishing blood traditional Chinese medicine XJCRTSZW can down-regulate the expression of autophagy related molecules LAMP2 and Cathepsin D to promote follicular development and repair damaged ovarian function. |
