The Confrontation Mechanism Of Guizhi Shaoyao Zhimu Standard Decoction On The Destruction Of Chondrocytes In Rats With Collagen-Induced Arthritis Model

1.Objective: Guizhi Shaoyao Zhimu Decoction is a famous and commonly used prescription for the treatment of rheumatoid arthritis in clinical practice,but its mechanism of action need further exploration。In this study,We established CIA(collagen induced arthritis)rat model,To investigate the mechanism of Guizhi Shaoyao Zhimu Decoction on joint inflammation and chondrocyte destruction in CIA model rats,so as to provide experimental basis for the clinical application of Guizhi Shaoyao Zhimu Decoction in the treatment of rheumatoid arthritis.2.Methods: We randomly divided 100 male Wistar rats into blank group and model group.Rats were induced to form collagen induced arthritis by subcutaneous injection of complete / incomplete Freund‘s adjuvant and mixed emulsion of bovine type II collagen.toinduced formation of CIA model.Rats which issuccessfully modeled were randomly divided into model group,methotrexate group,low-medium-high dose group of Guizhi Shaoyao Zhimu Standard Decoction group.The threshold of heat pain stimulation in hind feet of rats were measured regularly;TNF-α,IL-18,OPN and comp in rat serum were detected by ELISA;Pathological changes of ankle cartilage were observed in rats.Making statistical analysis of the above data.3.Results: 1)Guizhi Shaoyao Zhimu Standard Decoction can increase the threshold of thermal pain in CIA rats.2)Guizhi Shaoyao Zhimu Standard Decoction can reduce the level of TNF-α and IL-18 in serum of CIA rats.3)Guizhi Shaoyao Zhimu Standard Decoction can reduce the levels of OPN and comp in serum of CIA rats,which is significantly different from the model group,There is no significant difference compared with methotrexate group.4)The pathological results of cartilage tissue showed that the pathological score of the model group of CIA was higher than that of the normal group,and the pathological score of the treatment group was lower compare to the model group.Guizhi Shaoyao Zhimu Standard Decoction could alleviate the pathological changes of cartilage tissue.4.Summary: Guizhi Shaoyao Zhimu Standard Decoction can increase the threshold of thermal pain stimulation in CIA rats,which has better anti-inflammatory and analgesic effects.Guizhi Shaoyao Zhimu Standard Decoction may alleviate chronic inflammation of synoviumbyreducing the levels of inflammatory cytokines TNF-α and IL-18,to achieve antiinflammatory and analgesic effects.regulating RANKL / RANK / OPG,to intervene in the activation and maturation of osteoclast,intervene in the degradation of cartilage extracellular matrix,and prevent chondrocyte destruction.
