Effect Of Liyan Zhiyang Granule On Pulmonary Qi Deficiency Allergic Pharyngitis In Rats

Objective:In order to study the mechanism of Liyan Zhiyang Granules in the treatment of Allergic Pharyngitis(AP),this study used SD rats with Pulmonary Qi Deficiency type of AP model as the research object,and observed the number of mast cells(MC)in pharynx and thymosin lymphopoietin(TSLP),interleukin-4(IL-4),and Amphiregulin(AREG)expressions in serum of rats,to explore the role of Liyan Zhiyang Granules in treating AP rats with Pulmonary Qi Deficiency,providing theoretical basis for clinical application of Li Yan Zhi Yang Granules in treating AP.Method:1.55 male SD rats were selected and divided into cages according to the random weight method.They were set as group A(blank group),group B(blank control group),group C(model group),group D(model plus Liyan Zhiyang Granule group)and group E(model plus loratadine group),10 rats in each group.2.Establishment of Pulmonary Qi Deficiency type of AP rat model: by referring Yao Liu's smoking method,to replicated the pulmonary qi deficiency model to the rats in group B,C,D,E,after 4 weeks,to check if the rats have the syndrome of Pulmonary Qi Deficiency.After building the model of Pulmonary Qi Deficiency type on rats,the AP model should be started.There are two steps to build it.First,mixing ovalbumin(OVA)and aluminum hydroxide(AL(OH)3)in saline solution and injecting it into the intraperitoneal to do the basic sensitization;Second,using OVA to inject into the mucosa of the throat on rats.The basic sensitization should be performed for 2 weeks,and the throat injection should last for 10 days.After all the process of building the AP rats model,we will score the symptoms of these rats.3.Different treatment interventions were performed on the rats after modeling.Samples were collected from all rats after 2 weeks of treatment,and the distribution of MCin the pharynx was observed by amine blue staining.Enzyme-linked Immunoassay(ELISA)was used for detection of thymosin lymphopoietin(TSLP),interleukin-4(IL-4)and Amphiregulin(AREG).Result:After modeling,the AP rat model of Pulmonary Qi Deficiency type showed obvious pharyngeal symptoms such as scratching the mouth,cough,and outflow of pharyngeal secretions,and the behaviors of the rats in D and E groups observed the clinical symptoms were improved after different treatments.The number of pharyngeal mucosal MCs in group D and E decreased significantly,and there is no statistically significant difference between group D and group E(P = 0.526,P> 0.05).ELISA test showed that group E were lower than group D in the concentrations of TSLP,IL-4 and AREG in serum,while the concentrations of TSLP,IL-4 and AREG in serum of group D and E were lower than those of group C.Conclusion1.Liyan Zhiyang Granules can significantly improve the symptoms of AP rats with Pulmonary Qi Deficiency syndrome.2.Liyan Zhiyang Granules can reduce the distribution of mast cells in the pharyngeal mucosa and the concentrations of TSLP,IL-4 and AREG in the serum of AP rats with Pulmonary Qi Deficiency syndrome.3.The mechanism of Liyan Zhiyang Granules in treating Pulmonary Qi Deficiency AP may affect the activation of ILC2 s by reducing the number of mast cells and the expression level of TSLP,thereby limiting the production of cytokines such as IL-4 and AREG by ILC2 s.
