Study On The Relationship Between IL-10 Gene Polymorphism And EBV Infection In Children

Objective:To investigate the correlation between the polymorphism of loci-1082A/G,-819T/C and-592A/C in IL-10 and the susceptibility and severity of E BV infection among the children of the Han nationality in Hangzhou.Methods:A total of 260 children of Han nationality in Hangzhou who were hospitalized in the department of pediatrics of our hospital from January 2019 to December 2019 were collected and diagnosed as EBV infection by detection of four antibodies to EBV and real-time quantitative PCR detection of EBV-DNA.Among them,87 children with infectious mononucleosis((IM))and 173 children with previous EBV infection were diagnosed clinically.At the same time,78 children without EBV infection in pediatrics in our hospital were selected as negative control group.According to whether there was liver function impairment or not,all children with IM were divided into liver damage group(n=52)and non-liver damage group(n=35).After admission,the patients with previous history of IM and severe EBV infection,congenital malformation,severe organ dysfunction and malignant tumor,and those who had taken antineoplastic drugs,immunosuppressants and anti-tuberculosis drugs were excluded.All the children collected blood 1ml at one time as blood samples,and their family members signed the informed consent form before the examination,which was approved by the hospital ethics committee.DNA was extracted from blood samples and primers were designed with Primer 5.0.The PCR products were directly sequenced.After the information of-1082A/G,-819T/C and-592A/C sites of IL-10 gene was found in NCBI,the peak maps were compared with AB1 software,and the sequencing results were analyzed.The gene frequencies of the three loci were analyzed by SPSS25.0 software,and the gene polymorphism was compared by X2 test,P<0.05 indicates that there is a statistical difference.Result:1.The frequencies of AA,AG and GG genotypes of IL-10-1082A/G l oci in IM group were 90.8%,9.2%and 0.0%,and the frequencies of A and G alleles were 95.4%and 4.5%,In the previous infection group,the frequenci es of AA,AG and GG genotypes were 83.5%,15.0%and 1.2%,and the freq uencies of A and G alleles were 91.3%and 8.7%.In the negative control gro up,the genotypic frequencies of AA,AG and GG were 87.2%,12.8%and 0.0%,and the frequencies of A and G alleles were 93.6%and 6.4%,there was no significant difference in genotypes and alleles among the three groups(X~2=2.896?3.058,P>0.05),There was no significant difference between live r damage group and non-liver damage group in children with IM(P>0.05).2.The two loci of IL-10-819T/C and IL-10-592A/C are highly linked,the freq uencies of TT,TC and CC genotypes of-819T/C loci in IM group and AA,A C and CC genotypes of IL-10-592A/C loci were 50.6%,39.1%and 10.3%,the frequencies of T and C alleles at the-819T/C site and A and C alleles at the-592A/C site are 70.1%and 29.9%,in the previous infection group,the frequen cies of TT,TC,CC genotypes at-819T/C loci and AA,AC and CC genotypes at-592A/C loci were 48.0%,41.0%and 11.0%,the frequencies of T and C al leles at the-819T/C site and A and C alleles at the-592A/C site are 68.5%and31.5%,in the negative control group,the genotypic frequencies of 819T/C,A A,AC and CC at TT,TC,CC and-592A/C loci were 48.7%,42.3%and 9.0%,the frequencies of T and C alleles at the-819T/C site and A and C alleles at the-592A/C site are 69.9%and 30.1%,there was no significant difference in genotypes and alleles among the three groups(X~2=0.398?0.181,P>0.05).H owever,between the liver damage group and the non-liver damage group,the f requency of CC genotype of-819T/C site and CC genotype of-592A/C site in t he liver damage group was significantly higher than that in the non-liver dama ge group(X~2=9.795,P<0.05).There was no significant difference in allel es between the two groups(X~2=0.23,P>0.05).Conclusions:1.IL-10 gene polymorphism was found in children of Han nationality in Hangzhou area.2.The polymorphisms of-1082A/G,-819T/C and-592A/C in the promoter of IL-10 gene were not associated with EBV susceptibility.3.The polymorphism of-1082A/G locus was not associated with liver function impairment.4.The CC genotypes of-819T/C and-592A/C were correlated with liver function impairment in children with IM,CC genotype could increase the risk of liver function impairment in IM.5.The-819T/C genotype is highly linked to the-592A/C genotype.
