Chapter 1 Study on Changes of H2O2,b-FGF in Human Fallopian Tube Epithelial Cells after UU4 Infection Objectives By co-culture Ureaplasma urealyticum serotype 4?UU4?with human fallopian tube epithelial cells,the effects of different concentrations and different times of UU4 intervention on b-FGF and H2O2 contents of human fallopian tube epithelial cells were analyzed,and the relationship between H2O2,b-FGF and UU4 pathogenicity was discussed,so as to provide theoretical basis for the pathogenicity study of UU4.Methods 1 After the intervention of human fallopian tube epithelial cells with different concentrations of UU4 for 24 and 48 hours,the OD value of H2O2 in the superscript solution of cells in the control group and each experimental group was detected by ELISA,and the concentration of H2O2 was calculated,the changes of H2O2 in the superscript solution of cells in each group were compared,the experiment was repeated 3 times;2 After the intervention of human fallopian tube epithelial cells with different concentrations of UU4 for 24 and 48 hours,the OD value of b-FGF in the superfluid of cells in the control group and each experimental group was detected by ELISA,and the concentration of bFGF was calculated,the changes of b-FGF in the superfluid of cells in each group were compared,the experiment was repeated 3 times;3 After intervention of human fallopian tube epithelial cells with different concentrations of UU4 for 24 and 48 hours,the cell growth of the control group and the experimental group was detected by MTT colorimetry,and the H2O2,and with the increase of UU4 concentration and time,the expression of H2O2 increases,suggesting that produces toxic metabolite H2O2 may be one of the pathogenic mechanisms of UU4.2 UU4 can induce b-FGF production in human fallopian tube epithelial cells,and with the increasing UU4 concentration and time,the expression of b-FGF increases,suggesting that b-FGF may be involved in reducing UU4 damage to human fallopian tube epithelial cells.3 The H2O2 induced by UU4 was positively correlated with the expression of b-FGF in human fallopian tube epithelial cells.Chapter 2 Investigation of Mycoplasma Infection in Female Genital Tract Objectives To understand the female genital tract mycoplasma infection and drug resistance in this area,and explore the correlation between Ureaplasma urealyticum and Mycoplasma hominis infection and acute cervicitis.Methods 1006 female genital tract specimens were tested using mycoplasma culture,identification and drug sensitivity kits to analyze the female genital tract mycoplasma infection and drug resistance in this area.From 1006 patients,182 patients with confirmed acute cervicitis were selected as the case group.At the same time,186 non-acute cervicitis patients who were examined at the medical examination center of our hospital were selected as the control group.The detection rate of mycoplasma infection was compared between the two groups.Correlation between mycoplasma infection and acute cervicitis.Results 1 Among the 1006 samples,the total positive rate of mycoplasma was 51.7%?520/1006?,single Ureaplasma urealyticum?UU?infection was the most?40.76%?,followed by UU+MH mixed infection?8.55%?,and single Mycoplasma hominis?MH?infection was the least?2.39%?.Drug sensitivity results showed that the three types of mycoplasma infection had high sensitivity to josamycin,doxycycline and minocycline,and low sensitivity to quinolones and aminoglycosides,all of which had different degrees of drug resistance.The drug resistance rate of mixed UU+MH infection was higher than that of single UU and MH infection,and had multiple drug resistance.2 The UU detection rate in the case group was 35.16%?64/182?,and the UU detection rate in the control group was 23.66%?44/186?,the difference between the two groups was statistically significant??2 = 5.876,P<0.05?;The MH detection rate in the case group was 5.49%?10/182?,and the MH detection rate in the control group was 2.15%?4/186?,there was no significant difference between the two groups??2 =2.811,P>0.05?;The UU+MH detection rate in genital tract mycoplasma infection is mainly UU infection,MH mostly appears as mixed infection,josamycin,doxycycline,minocycline can be used as the first choice for treating mycoplasma infection in
