Mycoplasma is the smallest original nuclear microorganisms can grow on artificial medium, it is no cell, showed the highly polymorphic, can causes many diseases such as urogenital infection(urethritis、cervicitis), prostatitis, infertility, abortion, mucous purulent cervicitis. Antibiotic such as tetracycline class and macrolides class are the main effective drug to treat urogenital infection. Due to overuse of antibiotics, used incorrectly, and the appear of mycoplasma resistance genes, cause mycoplasma resistance enhanced, Thus lead to mycoplasma result disease curative effect not beautiful. Therefore, mycoplasma drug-resistant problem is urgentedly needed to solve. One of the effective ways to solve microbial resistance issue is to develop new antibiotic that different from to traditional antibiotics mechanism. Antibacterial peptides is a kind of small molecule polypeptide which induction produced in the body. Its antibacterial activity is effective, stability, and it can interact with the immune system. This study analyzed the infection situation of genitourinary tract mycoplasma and drug resistance in yangzhou region; Using PCR technique test the mycoplasma and drug resisitance gene. And extractde the restructuring antimicrobial peptides using genetic engineering technology for in vitro antibacterial experiment. The results of the study indicate that the infection rate of Uu was34.24%, the mixed infection rate of Uu and Mh was2.17%. the simple infection rate of Uu was3.26%, this result is consistent with other studies. Results show that the mycoplasma infection caused by Uu is the main in yang zhou region currently. And the same patients may also infect Uu and Mh. Antimicrobial susceptibility test results show that Ureaplasma urealyticum have different levels of resistance to nine kinds of antibiotics, this ratio is51.02%. highest sensitivity is clarithromycin, original amphotericin and delivery sand amphotericin (sensitive rate reached more than90%), so these three kinds of drugs are first choice for treat Uu and Mh. Withstand drug with the highest level are ciprofloxacin and methylpiperazine (withstand drug to a greater extent is more than72%), the results show that these drugs are not suitable for the the treatment of Uu and Mh. Meanwhile, this experiment finds that in368cases the positive proportion of mycoplasma is49.73%by genetic amplification, but the proportion of positive is38.85%by culture method. Gene amplification technologies can detect the mycoplasma DNA sensitive. Moreover, the results of gene amplification and classic antimicrobial susceptibility test are consistent, PCR technology has the advantages of simple and fast, and has great clinical application potential. In order to solve the mycoplasma resistance issue, This experiment does in vitro antibacterial experiment with restructuring antibacterial peptides for antibacterial activity of drug-resistant strains, the purpose is to solve the problem of clinical drug resistance. Results show that the certain concentration antibacterial peptides can inhibit the growth of drug-resistant strains. Using antimicrobial peptides can provide new ways to solve the future clinical resistance issue and provide direction for new drug research and development.
