Objective By observing the change of the course of rock salt aerosol on silicosis rats,the expression of inflammatory factors,the level of oxidative stress and the degree of fibrosis during the development of silicosis were detected,and its preventive and therapeutic effects on silicosis rats were explored,provides a natural,non-drug,safe and effective new method for the prevention and treatment of silicosis in the future.Methods SPF adult male SD rats were randomly divided into normal control group,rock salt control group,silicosis model group,rock salt prevention group and rock salt treatment group according to body weight.Each group of rats was divided into 7 subgroups according to the sampling time(4d,7d,14 d,28d,42 d,56d,and 84d),with 6 in each subgroup.Rats in the silicosis model group,rock salt prevention group,and rock salt treatment group were established by using the dynamic dust method(HOPEMED8050 series dynamic poison control device,the cabinet silicon dust concentration is 2000mg/m3,daily dyeing dust for 3 hours);rats in the rock salt prevention group and rock salt control group were received salt rock inhalation intervention from the time of dust exposure begin(ASA-01.3 dry salt aerosol treatment instrument,the mass concentration of rock salt in the treatment room was 20mg/m3,and treated for 30 minutes daily treatment);the rock salt treatment group rats were treated with rock salt from 14 d of dust exposure;the normal control group was not treated.The lung tissue at the maximum transverse diameter of the right lower lobe of the rat was taken for sectioning,and Hematoxylin-eosin(HE),Masson staining,and Sirius red staining were all used to observe the morphological and pathological changes of lung tissue in different groups of rats.The left lung tissue was prepared with ultrasonic tissue disruption instrument for homogenization of the lung tissue,and the enzyme-linked immunosorbent assay(ELISA)was used to detect the Surfactant protein D(SP-D),Clara cell protein 16(CC-16),Total antioxidant capacity(T-AOC)and Superoxide dismutase(SOD)changes.Taken the rat bronchus alveolar lavage fluid,and the total number of cells in the rat alveolar lavage fluid was counted by the cell counter trypan blue exclusion method.ELISA was used to detect the changes of Tumor necrosis factor-?(TNF-?),Interleukin-6(IL-6)and Transforming growth factor-?(TGF-?)in alveolar lavage fluid.Results 1 HE,Masson and Sirius scarlet staining results show that this experiment has successfully constructed the rat silicosis model.The lung tissue of the silicosis model group was the most severe,followed by the rock salt treatment group,and the rock salt prevention group was milder.2 The comparison of the total cell counts of bronchus alveolar lavage fluid in rats showed that the degree of change in the normal control group and the rock salt control group was not obvious.The silicosis model group increased significantly with the increase of dust exposure time,and the increase rate of the rock salt prevention group was slower than that of the silicosis model group.The growth rate of the rock salt treatment group was between that two groups.3 The levels of TNF-? in bronchus alveolar lavage fluid of rats in the silicosis model group,rock salt prevention group and rock salt treatment group all increased first and then decreased,but the silicosis module was always higher than the other two groups.4 The level of IL-6 in bronchus alveolar lavage fluid of rats in the rock salt prevention group was always higher than that of the control group,lower than that of the silicosis model group,and was significantly changed from the control group from 14 days and from the silicosis model group from 28 days.5 CC-16 levels in the lung tissue of silicosis model group,rock salt prevention group and rock salt treatment group rats showed a decreasing trend since dusting,and the silicosis model group was always lower than the other two groups.6 Examine the change of SP-D level in rat lung tissue,except for 84 days,the rock salt prevention group and rock salt treatment group were lower than the silicosis model group and higher than the control group.7 Except for 84 days,the levels of T-AOC and SOD in the lung tissue of the silicosis model group,rock salt prevention group and rock salt treatment group all showed a decreasing trend since dusting,but the T-AOC and SOD of the rock salt prevention group and rock salt treatment group were both higher than those of the silicosis model group at each time point.8 The levels of TGF-? in bronchus alveolar lavage fluid of rats in the silicosis model group,rock salt prevention group and rock salt treatment group all increased first and then decreased,but the silicosis module was always higher than the other two groups.Conclusions 1 Rock salt aerosol intervention can reduce the level of inflammation in the lung,regulate oxidative stress injury,and inhibit silicosis fibrosis.2 Early intervention of rock salt aerosols is more effective for delaying the pathogenesis of silicosis.Rock salt aerosol therapy may become one of the effective means of prevention and treatment of silicosis.Figure3;Table10;Reference 147...
