Objective This study explored the effects of marigold lutein on the proliferation and apoptosis of human breast cancer MCF-7 cells.And through the detection of the proliferation and apoptosis-related proteins Bax,Bcl-2 and ER expression,explore the mechanism of its effect on breast cancer apoptosis.Provide experimental basis for the clinical application of lutein.Methods Breast cancer MCF-7 cells in the logarithmic growth phase were randomly divided into six groups in vitro: the lutein treatment group and the control group were set up with time gradients for experiments.The CCK8 method was used to observe the effect of lutein on the proliferation of MCF-7 cells,and the normal growth curve of MCF-7 cells was drawn according to the absorbance value.According to the formula inhibition rate =(1-drug group A value / control group A value)× 100%,the cell growth curves were drawed at different times and different concentrations.The morphological changes of cells at various concentrations after lutein treatment were observed under an electron microscope.Flow cytometry was used to detect the effect of different concentrations of lutein on the apoptosis rate of MCF-7 cells.Immunoblotting technique was used to detect the changes of apoptosis-related protein Bax and Bcl-2 protein expression and the changes of estrogen receptor ER ? and ER ? protein expression in MCF-7 cells treated with different concentrations of lutein.Real-time PCR further detected the relative expression of apoptosis factors Bax and Bcl-2,proliferation factor Ki67,estrogen receptors ER ? and ER ? at different concentrations.Results Lutein significantly inhibited the proliferation of breast cancer MCF-7 cells,and showed a time-dependent and dose-dependent relationship(P <0.05).The IC50 value of lutein acting on MCF-7 cells at 48 h is 39.153?g / ml,so the optimal drug concentration is40?g / ml.Compared with the control group,MCF-7 cells treated with lutein showed obvious changes in cell morphology.The results of flow cytometry showed that lutein increased the apoptosis rate of MCF-7 cells and the apoptosis rate and lutein concentration showed a significant dose-dependent relationship(P <0.05).Western blot analysis showed that the expression of Bcl-2 protein and estrogen receptor ER ? in breast cancer MCF-7 cells was significantlydown-regulated after 48 hours of lutein treatment(P <0.05),and the expression of Bax protein and estrogen receptor ER ? significantly increased(P <0.05).And showed a clear dose dependence.Real-time PCR results showed that after 48 hours of lutein treatment,the expression of Bcl-2,Ki67 and ER ? m RNA in breast cancer MCF-7 cells decreased significantly(P <0.05),and the expression of Bax and ER?m RNA increased significantly(P<0.05).Conclusion Marigold lutein inhibited the proliferation of MCF-7 breast cancer cells and induced apoptosis.It may affect cell differentiation by regulating the expression of estrogen receptors ER? and ER?.
