Study On The Metabolomics Characteristics Of Serum In The Specific Endowment Constitution Based On LC–MS

Objective: To compare the changes of endogenous metabolites in serum of the specific endowment constitution and peace constitution based on LC-MS metabolomics research methods,to find the metabolomics characteristics of the specific endowment constitution,and to explore the basis for the classification of the constitution and formation mechanism.Methods: The 2009 edition of "Chinese Medicine Constitution classification criteria" and the inclusion and exclusion criteria of this topic were used to recruit 18 special constitution group and 20 Peace Constitution Group,the serum metabolites of the two groups were compared by non-targeted metabonomics technique,and the difference of metabolites and their changes between the two groups were found by multivariate statistical analysis,the potential metabonomics markers of the special constitution were identified and analyzed by metabolic database.Results: 1.In the PCA and PLS-DA model discrimination diagrams,the samples of the specific endowment constitution and peace constitution showed an aggregate distribution in the group and a significant separation between the groups.The results showed that there were differences in serum metabolites between the specific endowment constitution and peace constitution.2.The comparative study of serum metabolites in the specific endowment constitution and peace constitution found that there were significant differences in the 12 kinds of metabolites,with statistical significance(P<0.05).They were Arachidonic acid,Lauric acid,glyceric acid,L-histidine,tryptophan,serine,alanine acid,serotonin,epinephrine,histamine,Taurine,Citric acid;compared with the peace constitution,the specific endowment constitution is in Arachidonic acid,Lauric acid,L-histidine and histamine were significantly up-regulated and the other metabolites were significantly down-regulated,with statistical significance(P<0.05).3.The KEGG Pathway enrichment results showed that the four metabolic pathways in the specific endowment constitution were significantly different from those in the peace constitution(P<0.05).The metabolic pathways were: Taurine and hypotaurine metabolism,Arachidonic acid metabolism,Histidine metabolism,and Inflammatory mediator regulation of TRP channels.Conclusion: 1.Metabonomics research methods can be objective classification of physical fitness,compared with normal physical fitness,special physical fitness has significant differences in metabolites.2.There are differences in energy metabolism,fatty acid metabolism,amino acid metabolism,as well as changes in neurotransmitters,hormone regulation,etc.3.By studying the metabolomics characteristics of the specific endowment constitution,it is found that the prone to allergic diseases of the specific endowment constitution may be related to the existence of high inflammation levels and changes in immune function.
