Objective: To provide a scientific basis for the diagnosis and treatment of brain damage caused by kidney cold syndrome based on the viewpoint of "kidney and brain homology and same disease treatment ".Methods :160 female SD rats were randomly divided into two parts except normal control group.In part,hydrocortisone was used to irrigate the stomach for14 days to establish a model of kidney yang deficiency.After successful modeling,the randomized groups were as follows: kidney yang deficiency model group,yizhiren low dose group,yizhiren high dose group,yizhiren-5 low dose group,yizhiren-5 high dose group,compound danshen tablet positive control group,ferulei-8 mongolian drug positive control group.The other part was to establish a model of kidney cold(yang deficiency)syndrome after 50 days of gastric perfusion with the Mongolian medicine Huangbai and Haijinsha.After the model was successfully constructed,the rats were randomly divided into three groups: kidney cold(yang deficiency)syndrome model group,Yizhiren low-dose group,Yizhiren-5 low-dose group,Yizhiren-5 high-dose group,compound salvia miltiorrhiza tablet positive control group,ferula-8-monia positive control group.The body weight and body temperature of rats were measured weekly during administration,and blood rheology was measured and compared in each group of serum c AMP?c GMP?c AMP/c GMP?E2?cort?T4?17-OHCS?NE?TNF-a ?BA?IL-8?Renin?hcy,as well as GBAB?Gly?Ach?NO?Renin?LDH?Ang-2 in brain tissue samples.Results :1.The hydrocortisone modeling part :(1)After modeling,the body weight decreased compared with the normal control group,and the body temperature decreased slightly.Decreased intake and drinking water,decreased activity,slow response,decreased body temperature,decreased body mass,sparse body hair loss,chills,arch back and other symptoms in the model-making rats.(2)After modeling,compared with the normal control group,the NO of serum c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS?NE?BA? brain tissue in the model group of kidney yang deficiency were significantly decreased(p<0.01),and the GBAB?Gly?Ach?Renin?LDH?Ang-2 of serum and brain tissue and hemorheology were significantly improved(p<0.01).(3)Compared with the model group,the brain tissue NO c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS?NE?BA? Yizhiren low dose group was significantly improved(p<0.05),and the cut values in serum TNF-a ?IL-8?Renin?hcy? brain tissue GBAB?Gly?Renin?LDH?Ang-2 and hemorheology were significantly decreased(0.01).(4)Compared with the model group,the NO of brain tissue c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS?NE?BA?Yizhiren high-dose group was significantly improved(p<0.05),the GBAB?Gly?Ach?Renin?LDH?Ang-2 of serum and brain tissue and the high cut value of hemorheology were significantly decreased(p<0.01),and there was no significant difference in other indexes pp<0.05).(5)Compared with the model group,the brain tissue NO c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS?NE?BA? Ang-2 and hemorheology were significantly decreased(p<0.01).(8)Compared with the model group,the NO of brain tissue in the E2?cort?T4?17-OHCS?NE?BA? control group was significantly improved(p<0.05),the serum TNF-a ?IL-8?Renin?hcy? brain tissue GBAB?Gly?Ach?Renin and hemorheology were significantly decreased(0.01),and there was no significant difference in other indexes pp<0.05).(9)Compared with the normal control group,the serum c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS of hydrocortisone kidney yang deficiency model group decreased significantly and had obvious statistical significance(p<0.01).There was no significant difference in serum c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS in the model group of Mongolian medicine kidney cold(Yang deficiency)syndrome compared with hydrocortisone kidney yang deficiency model group(p>0.05),which met the evaluation criteria of kidney yang deficiency model.Huang Bai and Hai Jin Sha successfully established kidney cold(Yang deficiency)syndrome.2.Mongolian medicine Phellodendron chinensis and Haijin Sha modeling part :(1)after modeling,compared with the normal control group,weight loss,body temperature has a slight downward trend.Decreased intake and drinking water,decreased activity,slow response,decreased body temperature,decreased body mass,sparse body hair loss,chills,arch back and other symptoms in the model-making rats.(2)After modeling,compared with the normal control group,the NO of serum c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS?BA? brain tissue in the model group of kidney yang deficiency were significantly decreased(p<0.01),and the GBAB?Gly?Ach?Renin?LDH?Ang-2 of serum and brain tissue and hemorheology were significantly improved(p<0.01).(3)Compared with the model group,the brain tissue NO E2?cort?T4?17-OHCS? T4 ?17-OHCS of Yizhiren high dose group was significantly improved(p<0.05),the GBAB?Gly?Ach?Renin?LDH?Ang-2 values of serum TNF-a ?IL-8?Renin?hcy? brain tissue were significantly decreased(p<0.01),and there was no significant difference in other indexes pp<0.05).(5)Compared with the model group,the c AMP?c AMP/c GMP?E2?cort?BA of Yizhiren-5low dose group was significantly improved(p<0.05),the GBAB?Ach?Renin?LDH?Ang-2 values of serum TNF-a ?IL-8?Renin?hcy? brain tissue were significantly decreased(p<0.01),and there was no significant difference in other indexes pp<0.05).(6)Compared with the model group,the E2?cort?T4?17-OHCS?T4 of Yizhiren-5 high dose group was significantly improved(p<0.05),the GBAB?Ach?Renin?LDH?Ang-2 values of serum c GMP?TNF-a ?IL-8?Renin?hcy? brain tissue were significantly decreased(p<0.01),and there was no significant difference in other indexes pp<0.05).(7)Compared with the model group,the brain tissue NO c AMP?E2?cort? compound salvia miltiorrhiza tablet positive control group was significantly improved(p<0.05),and the serum TNF-a ?hcy? brain tissue GBAB?Ach?Renin?LDH?Ang-2 and hemorheology were significantly decreased(p<0.01).(8)Compared with the model group,the E2?cort of compound salvia miltiorrhiza tablet positive control group was significantly improved(p<0.05),and the serum TNF-a ?IL-8?Renin?hcy? brain tissue GBAB?Ach?Renin?LDH?Ang-2 and hemorheology were significantly decreased(p<0.01).conclusions :(1)in this experiment,the serum c AMP?c AMP/c GMP?E2?cort?T4 ? 17-OHCS of the model group of mongolian medicine kidney cold(yang deficiency)syndrome was significantly decreased compared with that of hydrocortisone kidney yang deficiency model group compared with that of hydrocortisone kidney yang deficiency model group in normal control group(p<0.01),which met the evaluation criteria of kidney yang deficiency model.hydrocortisone successfully established the model of kidney yang deficiency.There was no significant difference in serum c AMP?c AMP/c GMP?E2?cort?T4?17-OHCS in the model group of Mongolian medicine kidney cold(Yang deficiency)syndrome compared with hydrocortisone kidney yang deficiency model group(p>0.05),which met the evaluation criteria of kidney yang deficiency model.(2)Compared with the normal control group,there were significant changes in the clinical diagnostic TNF-a ?IL-8?Renin?hcy?GBAB?Gly?Ach?NO?Renin?LDH?Ang-2 and measurement of hemorheology of ischemic cerebrovascular disease in the model group of kidney cold syndrome,which confirmed that the kidney cold syndrome can lead to brain damage.(3)Using Yizhiren and Yizhiren-5 and other therapeutic drugs to elucidate the changes of biochemical indexes of ischemic cerebrovascular disease caused by kidney cold syndrome,it is proved that Yizhiren and Yizhiren-5 have significant prevention and treatment effect on brain damage caused by kidney cold syndrome. |
