| Object: To observe the effect of the compatibility of HT 7” and SP 6 on serum metabolites and their related pathways in patients with insomnia,in order to explore the mechanism of HT 7 and SP 6 compatibility in treatment of insomnia from the microscopic level of serum metabolites.Methods: 1.30 patients with insomnia were randomly divided into Shenmen group(n=10),Sanyinjiao group(n=10)and Shenmen-sanyinjiao group(n=10),and 10 healthy people were set as healthy group.Electro-acupuncture treatment on HT 7,SP 6,and both HT 7 and SP 6 were performed to corresponding groups,respectively.The patients in the three groups received two courses of electro-acupuncture therapy with dilatational wave,frequency of 5Hz/25 Hz.The patients were treated once a day persisted for 30 minutes for 5 consecutive days as one course,and received two courses of treatment which were 10 times in total.2.The venous blood samples were collected from patients with insomnia in Shenmen group,Sanyinjiao group and Shenmen-sanyinjiao group and healthy subjects at enrollment,then LC-MS/MS technique of untargeted metabolomics was performed to detect the metabolites in the serum of patients with insomnia before treatment as well as healthy people.Subsequently,the potential metabolic markers in patients with insomnia were screened.3.The venous blood samples were collected from patients with insomnia in Shenmen group,Sanyinjiao group and Shenmen-sanyinjiao group after treatment,and based on the identified potential metabolic markers of insomnia patients before treatment,the differential ones between the patients after treatment in shenmen-Sanyinjiao group and Shenmen group,and between the patients after treatment in shenmen-Sanyinjiao group and Sanyinjiao group were screened,and the pathway information of the differential metabolites were analyzed.Results: 1.Compared the three groups of insomnia patients with healthy group before treatment,32 potential metabolic markers were screened out,and the relative concentration of which was significantly upregulated compared with the healthy group.2.After treatment,16 of the 32 potential metabolic markers of insomnia patients were significant different between Shenmen-Sanyinjiao group and Shenmen group.(1)In the positive ion mode,14 potential biomarkers restored normal level in Shenmen-Sanyinjiao group after treatment,which were reduced,but still significantly different from healthy group in Shenmen group after treatment compared with that before treatment.(2)In the negative ion mode,2 potential biomarkers were reduced.And one metabolite PS(14:1(9Z)/14:1(9Z))of which had no significant differences with healthy group in Shenmen-Sanyinjiao group after treatment.The other one metabolite Schidigerasaponin B1 was reduced but still significantly different from healthy group in Shenmen group after treatment compared with that before treatment.3.After treatment,no significant differences were observed between Shenmen-Sanyinjiao group and Sanyinjiao group.24 of 32 potential biomarkers were identified with obvious change trend.(1)Among which 14 potential biomarkers in positive ion mode reduced to healthy level in Shenmen-Sanyinjiao group,and they slightly increased in Sanyinjiao group,which were higher than that in healthy group.(2)In the negative ion mode,10 potential biomarkers reduced notably in Shenmen-Sanyinjiao,and PS(14:1(9Z)/14:1(9Z))had no significant differences with healthy group and the other nine of which were still significantly different from that in healthy group after treatment.In Sanyinjiao group,the all 10 potential biomarkers reduced to healthy level.Conclusions: 1.The compatibility of acupoints of HT 7 and SP 6 has the advantage of synergistic effect over the single use of HT 7 or SP 6 in improving the level of serum metabolites in patients with insomnia.2.The advantage of synergistic effect of the compatibility of acupoints HT 7 and SP 6 may be related to reducing the level of glycerinphosphoserine metabolites and Schidigerasaponin B1. |
