Objective: To evaluate the therapeutic effect and mechanism of yangxinxue Granules on chronic heart failure(chronic heart failure CHF)rats by observing the effects of yangxinxue Granules on the indexes of color Doppler,ATP content of myocardial tissue,histopathological changes of heart and invasive hemodynamics.Research method:90 male SD rats were injected with adriamycin to induce CHF animal model.After 8 weeks,echocardiography was performed to evaluate whether the model was successful.The The surviving 75 rats with successful modeling were randomly divided into high dose group of Yangxinxue granules,middle dose group of Yangxinxue granules,low dose group of Yangxinxue granules,benazepril group and model group.Another 10 male SD rats without model were set as the blank group.Then the blank control group and the model group were given water by gavage;The high,middle and low dose groups of yangxinxue granules continued to be infused with the yangxinxue granules of different concentrations;Benazepril hydrochloride group was given the same of human body equivalent dose of benazepril hydrochloride solution by gavage.The drugs were given to the stomach every 24 hours,and the experiment was finished 3 weeks later.At the same time,during the three weeks of drug intervention,except for the blank group,all the rats in the other groups were given adriamycin(2mg/kg)once a week to maintain the heart function in the state of chronic heart failure;At the same time,the blank group was injected with the saline once a week.At the end of the experiment,the general condition of each group of rats was observed;the cardiac function indexes were evaluated by the means of echocardiography;the ATP content of myocardial tissue was measured by ATP content detection kit;the hemodynamic indexes of each group of rats were collected by multi-channel physiological signal acquisition system;the cardiac tissue was taken for HE staining and Masson staining for histomorphology observation.Research result:1.After 8 weeks of adriamycin,SV,CO,LVFE and LVFS in the model group were significantly lower than those in the blank group(P<0.01),CHF rat model was successfully induced.2.After 3 weeks of traditional Chinese medicine treatment,compared with the model group,the general situation of the rats in each dose of yangxinxue granule,such as spirit,activity,skin,hair,stool and ascites,was improved.3.After 3 weeks of traditional Chinese medicine treatment,SV,CO,LVEF and LVFS in the model group were significantly lower than those in the blank group(P<0.01);SV,CO,LVEF and LVFS in the high dose group of yangxinxue granules were significantly higher than those in the model group(P<0.01);SV,LVEF and LVFS in the middle dose group of yangxinxue granules were significantly higher than those in the model group(P<0.01),and CO in the middle dose group was significantly higher than those in the model group(P<0.05);The SV,CO,LVEF and LVFS in the low dose group of yangxinxue granules were higher than those in the model group,but the difference was not statistically significant(P>0.05).4.After 3 weeks of traditional Chinese medicine treatment,the content of ATP in myocardial tissue of model group was significantly lower than that of blank group(P < 0.05);the content of ATP in myocardial tissue of high and medium dose group of yangxinxue granules was significantly higher than that of model group(P < 0.01);the content of ATP in myocardial tissue of low dose group of yangxinxue granules was not significantly higher than that of model group(P>0.05).5.After 3 weeks of traditional Chinese medicine treatment,the LVSP and |±dp/dtmax| of the model group were significantly lower than those of the blank group(P<0.01),the LVEDP of the model group was significantly higher than that of the blank group(P<0.05),the|±dp/dtmax| of the middle dose group of yangxinxue granules was significantly higher than that of the model group(P<0.01),and the LVEDP of the middle dose group was significantly lower than that of the model group(P>0.05);Compared with the model group,the|±dp/dtmax| in the high and low dose groups of yangxinxue granules increased significantly(P<0.05),the LVEDP in the high dose group decreased significantly(P<0.01),and the LVEDP in the low dose group decreased significantly(P<0.05).Compared with the model group,the LVSP of the high,middle and low dose groups of yangxinxue granules increased to some extent,but the difference was not statistically significant(P>0.05);the heart rate of each treatment dose group of yangxinxue granules and the blank group and the model group did not change much,and the difference was not statistically significant(P>0.05).6.After 3 weeks of treatment with traditional Chinese medicine,the myocardial cells in model group were observed under HE staining,which showed that the transverse lines of myocardial cells were fuzzy,the cells were edematous,some of the cells were stained deeper,the cell volume was reduced,and vacuoles were found in the cells,the morphological changes of cell injury were more serious than those of each treatment dose group of yangxinxue granules.HE staining showed that the high dose group of yangxinxue granules had capillaries and hyperemia in the myocardial interstitium,and some of the myocardial cells were edematous.The morphological changes of myocardial cells in the high dose group were lighter than those in the model group,the middle and low dose groups of yangxinxue granules,and the overall morphological changes were lighter than those in the low,middle dose groups of traditional Chinese medicine and the model group.7.After 3 weeks of treatment with traditional Chinese medicine,Masson staining showed that the myofibrils of myocardial cells in the model group were stained unevenly,the number of intercellular collagen fibers was increased,and the interstitium was widened.The number of intercellular collagen fibers in the high,middle and low dose groups of yangxinxue granules was more than that in the blank group,but less than that in the model group.There was no significant difference in the number of collagen fibers between the groups with high,medium and low doses of yangxinxue granules.Research conclusion:This study confirmed that yangxinxue granules can effectively improve the general state of CHF rats,improve the cardiac function,invasive hemodynamic indexes,increase the content of ATP in myocardial tissue and slow down the process of myocardial pathomorphological injury.The mechanism of yangxinxue granule in the treatment of chronic heart failure may be achieved by enhancing the cardiac contractility,increasing the ATP content of cardiac tissue and slowing down the process of myocardial pathomorphological injury.
