Objective: To investigate the effects of Xiaoyaosan and Lianpuyin on lipid-lowering,behavioral and metabonomics in NAFLD rats by intervening the model of liver depression and spleen deficiency and dampness and heat syndrome respectively,and comparing with atorvastatin.Methods: Fifty-four rats were randomly divided into four groups: normal control group(fed with common feed),NAFLD group(fed with high-fat emulsion),NAFLD damp heat group(fed with high-fat emulsion + artificial climate box for 12h/D),NAFLD liver depression and spleen deficiency group(fed with high-fat emulsion plus binding for 3h + tail clamping for 1h/D).After the completion of the model,the rats were divided into normal control group for drug intervention experiment NAFLD group,NAFLD damp heat syndrome control group,NAFLD liver depression and spleen deficiency syndrome control group,NAFLD Lianpuyin group,NAFLD Xiaoyaosan group,NAFLD western medicine group.After 14 days of drug intervention,behavioral experiments were carried out in rats.On the 15 th day of intervention,the serum of rats was dissected to detect TG,CHOL and GLU,the liver tissue was stained with he,and the liver,brain,colon tissue and feces were analyzed by non targeted metabonomics.Results: ? After 32 days of modeling,HE staining showed diffuse vacuoles with inflammation,less big bubbles and more small bubbles in the liver;Rats in the syndrome group of liver depression and spleen deficiency had squinting,and their stools were mostly unformed,or dried first and then thin,or dried at the same time,showing some symptoms of the syndrome;rats in the damp heat syndrome group had yellow,loose and rough hair,dark and lusterless,and a few stools had a little mucus Those who gradually see the perianal filth,redness and swelling,red and loose scrotal skin,showing some symptoms of damp heat syndrome.After the drug intervention experiment,the HE staining results of the liver in the model group showed fatty lesions,and the steatosis level of the liver cells in the Xiaoyaosan group decreased significantly,while the serum test results of the rats showed no significant difference.(2)In the open field experiment,there was no statistical significance(P> 0.05).In the model score of rats after drug intervention.In the elevated cross maze experiment,the model score of liver depression and spleen deficiency rats treated with Xiaoyaosan was significantly lower than that of the control group(P <0.01).(3)In metabonomics,berberine,1-arachidonol-sn-glycerin-3-phosphate choline,l-?-glycerophosphate choline,hypoxanthine,stachydrine and nicotinamide in the metabolites of Lianpuyin were significantly increased(FC < 0.1,P < 0.05).After the intervention of Xiaoyaosan,meprazole,2-(6-hydroxy-1h-indole-3-yl),acetamide,tirisolol,6-hydroxy-5-methyl-4,11-dioxodedecanoic acid,dodecasulfate,3-hydroxyglycyrrhetinic acid,5 '-deoxyadenosine,(2S,3S)-2,3-dihydro-3-hydroxy-o-aminobenzoic acid zwitterions were significantly reduced(FC > 2,P < 0.1).Conclusion: According to the changes of liver histopathology and the characteristics of rat body surface,it is suggested that the TCM syndrome model of rats is constructed successfully.On this basis,it is found that Xiaoyao Powder has a certain effect on improving liver fat pathological changes in rats with liver depression and spleen deficiency syndrome,and through behavioral experiments,it has a certain effect on relieving anxiety and antidepressant in rats with liver depression and spleen deficiency syndrome,and there is a big difference between the experimental group and other groups in the analysis of metabolites;it is found that Lianpu Yin has the effect of improving damp and heat symptoms in rats,and it is in the generation of In the analysis of metabolites,we found that there was significant difference between berberine and other metabolites in lianpuyin group and the control group,which may be related to the improvement of symptoms of damp heat syndrome.
