For the diagnosis and prognosis of many diseases,such as reproductive system disorders,blood diseases,organ diseases and so on,the using of advanced urine sample analysis equipment to quantify some of the diagnostically valuable components in urine samples is a necessary step.The test result of the constituents in the urine sample is one of the necessary basis for the doctor to make a correct diagnosis.Furthermore,urine testing is different from blood testing.Urine testing is simple,non-invasive,and has obvious advantages.The detection of the number of red blood cells and white blood cells in urine samples is the key parameter for diseases such as nephritis and urinary infection.Detecting the presence of red blood cells and white blood cells and the number of them in urine is an important indicator for diagnosing the disease and the extent of the disease.The methods for counting red blood cells and white blood cells in urine can be divided into two categories: automatic methods and manual methods.Among them,the automatic method is mainly realized by using automatic urine analyzer and other equipment,and the identification and counting of red and white blood cells mainly use image processing technology.This type of equipment has a large throughput,and it is expensive,so it is not suitable for small and medium-sized medical institutions.The artificial method is an another method of identifying and counting red blood cells and white blood cells in urine.This method is often used for diagnosis and observation of special patient samples.Microscopy is commonly used in clinical practice,but this method requires mechanical scanning.The throughout is much lower,and the sample will be disturbed during the mechanical scanning.At the same time,the counting person need to be highly concentrated during the counting process.Here,this paper studied the use of Fourier Ptychographic microscopy(FPM)to solve the problems in manual counting methods,and considered the impact of environmental disturbances on urine white blood cell counts.This microscopic imaging method can synthesize a series of lower-resolution,wide-field-of-view images in the Fourier domain with high-resolution,wide-field-of-view images,so this method can avoid the trouble and error of mechanical scanning.Compared with traditional microscope devices,FPM uses an LED array as a light source and uses a CCD to capture images.The study verified the reliability of FPM in counting cells by performing two sets of comparative experiments,one was cells counting based on FPM of deviation vector correction and symmetrical lighting,and another was automatic urine analyzer counting.On the basis of ensuring the accuracy of the red blood cell and the white blood cell counting of the sample image,the FPM of the vector correction and symmetric lighting scheme improved the visual observation effect of the image.At the same time,the idea and the method of automatic counting with Binary Large Object analysis were given,and the subjective and objective evaluation methods were used to evaluate the image quality,which verified its effectiveness in the automatic counting ofred blood cells and white blood cells in urine sample images.The experimental results show that the counting accuracy of the method in this paper is comparable to that of KY906 D automatic urine sediment analyzer,which is about 95%.
