Snakes are a general term for snake subordinates,which belongs to the class of reptiles.They are a critical part of biodiversity,and they play an essential role in nature.They have a long evolutionary history,extremely diverse species,and extremely geographical distribution.Some snakes can also secrete venoms.They acquired a weapon called toxin in the course of their evolution,which is used to prey.This is also one of their important defenses.Snake venom is also quite diverse and complex with the evolution of snakes,Snake venom is a natural treasure house of bioactive peptides and proteins,with natural advantages in drug development and disease treatment.The main research object of this study is bungarotoxins(BGT).Bungarus multicinctus is the fourth most virulent snake on land and the most toxic snake in China.B.multicinctus venom is the solely peptide molecule listed as a potential toxin fighting agent among vertebrates in the world.It is also one of the poisonous snakes with the highest fatality rate in China.B.multicinctus venom is belong to neurotoxin,including a-bungarotoxin,?-bungarotoxin,y-bungarotoxin,?-bungarotoxin.?-bungarotoxin is the most abundant of the four toxins,belonging to presynaptic neurotoxin,with phospholipase A2 activity.The other three toxins belong to the post-synaptic neurotoxin,and belong to the three-finger toxin family.Up to now,the B.multicinctus venom is listed on the international red list of biological weapons.In order to deal with the potential threat of biological weapons,it is necessary to study the different components of venoms,establish the detection methods for different toxins,compare the toxicity differences and evaluate the protection and neutralization ability of antibodies.This study mainly focused on ?-bungarotoxin.First,the separation and purification process of ?-bungarotoxin was constructed,and its median of the lethal dose(LD50)was measured through mice experiments.Its toxicity was compared with other separated components.It is determined that ?-bungarotoxin is the most toxic toxin among them.After that,specific antibodies of different toxins were prepared respectively,compared with commercial a B.multicinctus antivenin purchased in China through the neutralization protection experiment,and conducted cross-neutralization experiment with top 9 snakes in our country.We found that ?-bungarotoxin antibodies and ?-bungarotoxin can specifically detect B.multicinctus venom.In addition,we also found that B.multicinctus antivenin has a certain neutralizing effect on Ophiophagus Hannah venom,but it has no protective effect on Bungarus fasciatus venom and Naja atra venom.It has certain guiding significance for the application of antivenin in clinical.
