Objective To investigate the relationship between the content of 3-bromotyrosine and asthma in asthmatic mice.Methods Forty-eight BABL / C female mice were randomly divided into an experimental group and a control group,with 24 mice in each group.The mice in the experimental group were sensitized by intraperitoneal injection of ovalbumin(OVA)combined with nebulized inhalation sensitizing solution(0.2 m L of sensitizing solution containing chicken egg protein 800 ug and aluminum hydroxide 1mg),induced the establishment of eosinophilic asthma mouse model,the control group of mice were intraperitoneally injected with the same amount of phosphate buffered saline(PBS)combined with inhalation 0.2 ml PBS build model.Four weeks later,the two groups of mice were sacrificed,and the bronchoalveolar lavage fluid(BALF)were collected for cell counting and classification.The content of 3-bromotyrosine in the urine and alveolar lavage fluid of the two groups of mice was detected by enzyme-linked immunosorbent assay,and compared and analyzed.HE staining was used to observe airway inflammation,airway remodeling and airway mucus secretion in lung tissue pathological sections.Results During the course of sensitization and stimulation,the mice in the experimental group showed asthma attacks.By the end of the sensitization and stimulation process,neither group of mice had died.The content of 3-bromotyrosine in alveolar lavage fluid and urine of the experimental group was significantly higher than that of the control group,with statistically significant differences(t=4.25,4.56,P?0.05).The content of3-bromotyrosine in alveolar lavage solution and urine of the experimental group was positively correlated with the number of eosinophils in alveolar lavage solution(r =0.89,0.46,P? 0.05).Conclusion 3-Bromotyrosine is highly expressed in alveolar lavage fluid and urine of asthmatic mice,which is closely related to airway inflammation,high secretion of airway and airway remodeling in asthmatic mice.Therefore,by detecting the level of 3-bromotyrosine in alveolar lavage fluid and urine,it may be helpful for the diagnosis of asthma.
