Background: Cervical cancer is a common gynecological malignant tumor,in the prevention and treatment of cervical cancer attention need to be paid.It is crucial for the early detection of cancer.At present,the screening methods we rely on are the follow-up after the formation of cancer,there is a lack of detection of biological factors that play a role in the formation of cancer.Tumor-associated protein Carbonic anhydrase ?(CA?)has two major forms,One is a cell-associated transmembrane protein seen on Western blots as a twin band of 54/58 kDa.The other is a soluble ptotein s-CA? of 50/54 kDa,Which is released into the body fluids most likely by proteolytic cleavage of the extracellular part from transmembrane and intracellular sequences.this protein primarily involved in maintaining the extracellular PH and overexpressed in a variety of tumors including cervical cancer.Methods: One hundred and sixty nine serum samples from one hundred seventy patients with cervical cancer and precancerous leisions were analyzed.For fifty two of seventy seven cervical cancer patients who underwent radical hysterectomy,samples were prospectively collected at 2 time points :before surgery and 7 days afterwards.Serum CA? was quantifified by ELISA.The serum CA9 concentration of cervical cancer patients and serum concentration of precancerous patients were statistically analyzed.The correlation between serum level and clinicopathological factors in cervical cancer patients was analyzed.At the same time,serum CA9 concentration was analyzed before and after operation,the correlation between serum CA9 concentration and serum SCCA concentration was analyzed.Westerblot was performed on the tissue samples of 49 cases of cervical cancer treated by radical surgery,and the correlation factors between CA9 expression and clinicopathological factors were analyzed.Results: Compared with the precancerous leisions group,serum levels of CA9 of cervical cancer group increased obvariously(P<0.01).Serum levels of CA9 have no relationship with age,pathological type,tumor staging and lymph node metastasis(P>0.05),but related to tumor diamater(P<0.05).Serum concentration of CA? showed meaningfully changes after surgery(P<0.05)The expression level of CA9 in tumor tissue is much higher than its counterpart in control group(P<0.01),Tumor CA9 levels have relationship with tumor staging and lymph node metastasis(P<0.05).Conclusion : CA9 ? is upregulated in cervical cancer compared with the precancerous leisions group.Serum concentration of CA? related to tumor diamater and has no relationship with pathological type,in this way CA9 could be used widely in the treatment of precancerous and detecting early cancer.CA? serum levels change meaningfully after surgery.In tumor tissue level,CA9 levels are related to tumor staging and lymph node metastasis.As biological factor related to cervical cancer,the mechanism and metabolic pathway of CA9 in the formation and development of cervical cancer remain to be further explored.
