BackgroundAllergic Rhinitis(AR)is a Noninfectious chronic disease(NCD).Although the incidence of AR increases year by year and has become a global problem,its pathogenesis has not been thoroughly studied so far.Previous studies suggested that the CAII gene(Carbonic anhydrase?,CA?)and CA? gene(Carbonic anhydrase ?,CA?)in the Carbonic anhydrase(CA)family were differently expressed in the nasal mucosa of AR patients and non-AR patients.Therefore,in order to further understand the role of CA in the pathogenesis of AR,we studied the expression of CA? and CA? from the lever of gene and protein,providing new ideas and theoretical basis for the mechanism research ofAR.MethodsIn this study,the inferior turbinate mucosal tissues of 25 non-AR patients became as the control groups,and inferior turbinate mucosal tissues of 19 AR patients became as the experimental groups.The inferior turbinate mucosa tissues were detected by real-time quantitative PCR,immunofluorescence staining and immunohistochemical staining from level of mRNA and protein of CA? and CA?.It has been reported that hCAII and hCA? are mainly distributed in submucosal glands,little in epithelial tissues.To get a better idea of how they were distributed,this research used the brush to remove nasal turbinate mucosa epithelial cells,nasal epithelial cells of 26 patients without AR became as control groups,nasal epithelium cells of 28 patients with AR became as the experimental groups,the nasal epithelium cells were detected by real-time quantitative PCR from the lever of mRNA.SPSS20.0 software was used for statistical analysis of all experimental data,and t-test of two independent samples was used to analyze dates.P<0.05 indicated that there were a statistical difference.Results1)By real-time fluorescent quantitative PCR,it found that the expression of hCA? gene was lower in inferior turbinate mucosal tissues of AR patients than the control groups,showing a statistical difference(P=0.020,P<0.05),while there were no significant differences in the expression of hCA? gene in nasal epithelial cells of AR patients between with the control groups(P=0.310).The expression of hCA? gene in AR patients' inferior turbinate mucosal tissues and AR patients' nasal epithelial cells showed no significant differences compared with the normal groups(P=0.139,P=0.911).2)By immunofluorescence staining,it showed that hCA? protein and hCA? protein were mainly distributed in the cytoplasm of the submucosal glands,the expressions of hCA?protein and hCA? protein were lower in inferior turbinate mucosal tissues of AR patients than patients without AR,showing a statistical difference(P=0.000,P=0.047).3)By immunohistochemical staining,it showed that hCAII protein and hCA? protein were mainly distributed in submucosal glands and were weakly expressed in AR patients but strongly expressed in non-AR patients.The expressions of hCAII protein and hCA? protein were significantly lower in inferior turbinate mucosal tissues of AR patients than that of the control groups,with statistical significances(P=0.000,P=0.000).ConclusionIn this study,hCA? and hCA? may have abnormal expression in AR patients' inferior turbinate mucosa from the level of gene and protein.hCA? and hCA? are mainly distributed in the cytoplasm of submucosal gland cells,with low expression in epithelial tissues.From the protein level and gene level,hCA? expression was lower in AR inferior turbinate mucosa,indicating that the lower expression of hCAII was closely related to the occurrence and development of AR.The expression of hCA? gene in AR inferior turbinate mucosa was not different from that in the non-AR group,but the protein level showed lower expression in AR inferior turbinate mucosa,so its role in AR needs further study.
